HMGB1/TLR4在卵巢癌中的表达及其与临床病理特征的关系 被引量:10

Expression of HMGB1/TLR4 in ovarian cancer and the relationships with clinicopathological features

摘要 目的探讨高迁移率族蛋白B1(HMGB1)、Toll样受体4(TLR4)信号蛋白在卵巢癌组织中的表达差异及其与临床病理特征的关系。方法采用RT-PCR和蛋白质印迹(Western-blotting)方法分别检测HMGB1、TLR4、核因子κB(NF-κB)及肿瘤坏死因子α(TNF-α)在卵巢癌组织及卵巢囊肿组织中的表达情况。结果逆转录聚合酶链反应(RT-PCR)检测显示HMGB1、TLR4、NF-κB、TNF-αmRNA在卵巢囊肿及卵巢癌组织中均有表达,卵巢癌组织中表达明显高于卵巢囊肿组,两组之间经t检验存在显著统计学意义(P<0.001)。Western-blotting检测显示:HMGB1、TLR4、NF-κB及TNF-α在卵巢囊肿及卵巢癌组织中均有表达,卵巢癌组织中表达明显高于卵巢囊肿组,差异有显著统计学差异(P<0.001),与实时荧光定量PCR结果相符。将卵巢癌患者根据临床分期、分化程度分组进行比较HMGB1、TLR4、NF-κB及TNF-α的表达差异,结果显示HMGB1、TLR4、NF-κB及TNF-α表达与临床分期和分化程度相关,分期较高的表达值较分期较低的表达明显增加,中低分化较中高分化的表达明显增加,差异具有统计学意义(P<0.001)。Pearson相关性分析显示,NF-κB、TNF-α作为下游因子与HMGB1、TLR4呈高度正相关(P<0.001)。结论 HMGB1、TLR4信号蛋白在卵巢癌组织中呈高表达并与临床病理特征相关,提示HMGB1、TLR4介导的炎性信号可能涉及卵巢癌发病机制。 Objective To explore the expression differences of high-mobility group box 1(HMGB1) and Toll-like receptor 4(TLR4)in ovarian cancer tissues and the relationships with clinicopathological features. Methods RT-PCR and Western blotting were used to detect the expression levels of HMGB1,TLR4,NF-κB,and tumor necrosis factor α(TNF-α) in epithelial ovarian cancer tissues and ovarian cyst tissues. Results RT-PCR showed that HMGB1,TLR4,NF-κB,and TNF-α mRNA expressed in epithelial ovarian cancer tissues and ovarian cyst tissues,the expression levels of HMGB1,TLR4,NF-κB,and TNF-α mRNA in epithelial ovarian cancer tissues were statistically significantly higher than those in ovarian cyst tissues(P〈0. 001). Western blotting showed that HMGB1,TLR4,NF-κB,and TNF-α expressed in epithelial ovarian cancer tissues and ovarian cyst tissues,the expression levels of HMGB1,TLR4,NF-κB,and TNF-α in epithelial ovarian cancer tissues were statistically significantly higher than those in ovarian cyst tissues(P〈0. 001). The results of RT-PCR and Western blotting were the same. The patients with ovarian cancer were divided into different groups according to clinical stages and differentiation degrees,the results showed that the expression levels of HMGB1,TLR4,NF-κB,and TNF-α were correlated with clinical stages and differentiation degrees. The expression levels of HMGB1,TLR4,NF-κB,and TNF-α in patients with high stage of ovarian cancer were statistically significantly higher than those in patients with low stage of ovarian cancer. The expression levels of HMGB1,TLR4,NF-κB,and TNF-α in patients with ovarian cancer of low and moderate differentiation were statistically significantly higher than those in patients with ovarian cancer of moderate and high differentiation(P〈0. 001). Pearson correlation analysis showed that NF-κB and TNF-αwere positively correlated with HMGB1 and TLR4 highly as downstream factors. Conclusion The expression levels of HMGB1 and TLR4 signal proteins in ovarian cancer tissues are high,which are correlated with clinicopathological features of ovarian cancer,which indicate that HMGB1 and TLR4-mediated inflammatory signals may be involved in the pathogenesis of ovarian cancer.
