摘要
为对提取工艺条件进行合理优化,采用微波辅助法提取结球菊苣总苷,并利用响应曲面法设计试验。在单因素试验的基础上,分别研究了乙醇体积分数、料液比、提取时间和提取温度对结球菊苣总苷提取率的影响。基于单因素试验结果,响应曲面法分析得到总苷提取的最优条件:乙醇体积分数66%,料液比1︰32(g/m L),提取时间17min,提取温度77℃,在此条件下,结球菊苣总苷的提取率为6.139%,与模型高度拟合。试验为从结球菊苣中提取分离总苷类化合物以及对结球菊苣总苷的开发利用提供了理论依据。
Response surface method(RSM) used to design and optimize the condition of the technological process,it mainly focused on microwave extraction of total glycosides from Cichorium intybus L..Based on the single-factor experiment,comprehensive influence of 4 factors contributing to the yield of total glycosides was inspected respectively:concentration of ethanol,solid-liquid ratio,extraction time and extraction temperature.Based on the results of the single factor test,results showed that the optimal values of the above parameters were determined as follows:concentration of ethanol was 66%,solidliquid ratio was 1︰32(g/m L),extraction time was 17 min,and extraction temperature was 77 ℃.Under the optimized conditions,the experimental value of yield of crude total glycosides was 6.139% and was in good agreement with the predicted value.The research offers a practical basis for the extraction of total glycosides from Cichorium intybus L.,and the development as well as utilization of the total glycosides.
