摘要
目的探讨中波紫外线UVB照射对皮肤成纤维细胞纤维连接蛋白(fibronectin,FN)及其选择性剪接片段EDA、EDB的影响。方法用RT-q PCR法检测UVB照射后成纤维细胞各时相FN及其EDA、EDB片段的mRNA表达水平。结果 UVB100m J/cm2照射后成纤维细胞FN及FN-EDA、EDB的mRNA表达均有下降。照射后1、2、4、8、12、24h组与对照组(0h)比较,差异有统计学意义(P均<0.05)。结论中波紫外线UVB下调皮肤成纤维细胞FN及FN-EDA、EDB mRNA表达,在UVB引起的皮肤成纤维细胞急性光损伤过程中,FN及FN-EDA、EDB可能发挥着重要作用。
Objective To evaluate the effects of UVB irradiation on the mRNA expression of fibronectin(FN)and alternatively spliced segments(EDA/EDB)in human dermal fibroblasts. Methods RT-qPCR was used to detect mRNA expression of FN and FN-EDA/EDB in UVB irradiated fibroblasts over a range of time points. Results Following exposure to a dose of 100mJ/cm2 of UVB irradiation, the mRNA levels of FN, EDA and EDB decreased as compared to unirradiated control. Significant statistical differences(P〈0.05)in FN, EDA and EDB mRNA expression were observed between the unirradiated control(0h)and irradiated groups(1,2,4,8,12,and 24h). Conclusion UVB is an effective factor to down regulate the mRNA expression of FN, EDA and EDB in human dermal fibroblasts. FN,EDA and EDB may play important roles in the process of UVB-induced acute photo-damage of human dermal fibroblasts.
