参苓白术散治疗脾虚泄泻模型小鼠肠道局部免疫机制研究 被引量:28

The local immune regulation of shen-ling-bai-zhu powder in traditional Chinese medicine spleen deficiency diarrhea model

摘要 目的探讨参苓白术散治疗脾虚泄泻模型小鼠肠道局部免疫机制。方法SPF级昆明小鼠40只按随机对照的原则分为:A组(正常对照组)、B组(模型组)、C组(参苓白术散治疗组)、D组(思密达阳性对照组),每组10只。计算每组小鼠脾/胸腺指数,光镜检测小鼠肠微绒毛和固有层的病理变化,用ELISA法检测小鼠血清IL-2、IL-12、TNF-α及肠组织s Ig A水平。结果模型组脾/胸腺指数显著降低(P<0.05),小鼠肠上皮固有层、微绒毛损伤明显,血清IL-2、IL-12、TNF-α及肠组织s Ig A水平显著下调(P<0.05),参苓白术散治疗组以上各种指标较脾虚模型组显著恢复。结论参苓白术散能够显著升高脾虚模型组小鼠脾/胸腺指数,上调其血清IL-2、IL-12、TNF-α及肠组织s Ig A水平,增强小鼠免疫功能,进而修复小鼠肠黏膜损伤,达到治疗脾虚泄泻的作用。 This study was designed to investigate the local immune regulation of shen-ling-bai-zhu powder intraditional Chinese medicine"spleen deficiency diarrhea"mice. Total of 40 SPF Kunming mice were randomlydivided into four groups: group A(control group), group B(model group), group C(shen-ling-bai-zhu powder group)and group D(positive control). Then the spleen and thymus indexes were calculated, the pathological changes ofintestinal mucosa were checked by light microscope, and the serum levels of IL-2, IL-12, TNF-α and s Ig A weredetected by ELISA. Data showed that in the model group, the spleen and thymus indexes were significantly reduced(P〈0.05), the intestinal epithelial mucosa and villus damaged significantly, and the levels of IL-2, IL-12, TNF-αand s Ig A were markedly downregulated, while all indicators above in Shen-ling-bai-zhu powder group weresignificantly recovered. These results suggests that Shen-ling-bai-zhu powder significantly increases spleen andthymus indexes in mice with spleen-deficiency diarrhea, and upregulates the levels of serum IL-2, IL-12, TNF-αand s Ig A in intestinal tissue. So the powder improves the immune function and repairs the intestine mucosal injuryof"spleen deficiency diarrhea"mice.
