摘要
目的了解广州市部分地区捕捉的小型兽类汉坦病毒感染情况,分析汉坦病毒基因型别。方法于2016年捕获广州市白云区、海珠区、花都区、从化区、增城区等5区的小型兽类,提取其肺组织样本RNA,采用特异性引物进行汉坦病毒的PCR检测,确定基因型。结果本研究共捕获小型兽类330只,其中褐家鼠195只(59.09%)、黄胸鼠62只(18.79%)、臭鼩鼱24只(7.27%)、板齿鼠16只(4.85%)、小家鼠14只(4.24%)、黑家鼠19只(5.76%)。经汉坦病毒特异性PCR检测发现其阳性率为6.06%(20/330),其中褐家鼠13份,黄胸鼠2份,板齿鼠2份,小家鼠1份,黑家鼠2份,不同动物间汉坦病毒阳性率差异无统计学意义(P>0.05),优势鼠种为褐家鼠;不同区间汉坦病毒阳性率差异有统计学意义(P<0.05),以海珠区阳性率最高。扩增阳性样本经核酸序列测定分析表明,所有基因型均为首尔型。结论广州地区小型兽类携带的汉坦病毒为首尔型,其中褐家鼠为主要的带毒兽类,海珠区小型兽类汉坦病毒阳性感染最为严重。
Objective To understand hantavirus infections of small mammals captured from Guangzhou city and analyzehantavirus genotypes and gene characteristics. Methods In 2016,RNA was collected from the lungs of small mammals in5 areas of Guangzhou(Baiyun area,Haizhu area,Huadu area,Conghua area,and Zengcheng area)and specific primerswere used to determine the genotype of hantaan virus L gene. Results A total of 330 small mammals were captured,including Rattus norvegicus(195,59.09%),Rattus flavipectus(62,18.79%),Suncus murinus(24,7.27%),Bandicota indica(16,4.85%),Mus musculus(14,4.24%),Rattus rattus(19,5.76%). 20 specimens were detected positive ofhantavirus. 13 were Rattus norvegicus,2 were Rattus flavipectus,2 were Bandicota indica,1 was Mus musculus,2 were Rattus rattus. The positive rate of hantavirus infection was 6.06%. There was no statistically significant difference in thepositive rate of hantavirus among different species(P〉0.05). The dominant rat species was Rattus norvegicus. The positiverate of hantavirus in each interval was statistically different(P〈0.05),and the positive rate in Haizhu area was the highest.All positive specimens were tested for nucleic acids,sequenced and analyzed,and their genotypes were Seoul type.Conclusion Hantavirus which is prevalent among rats in Guangzhou bearing the Seoul genotype. The dominant infectionrat species was Rattus norvegicus. The infection of small mammals in Haizhu area is the most serious.
