桃仁-大黄药对活血化瘀最佳配比的古代文献数据挖掘及其对寒凝血瘀大鼠模型的影响分析 被引量:6

Ancient literature data mining on the best ratio of Taoren-Dahuang to promoting blood circulation and removing blood stasis and its effects analysis on rat model of cold congelation and blood stasis

摘要 目的:挖掘古代文献数据中桃仁-大黄药对活血化瘀最佳配比和分析其对寒凝血瘀大鼠模型的影响。方法:(1)采用挖掘中医古今应用桃仁-大黄药对活血化瘀处方中的配比数据;(2)皮下注射肾上腺素加冰水浴法复制急性寒凝血瘀大鼠模型,设置桃仁-大黄药对(0∶1、1∶5、2∶5、2∶3、1∶1、3∶2、5∶2、5∶1、1∶0)9种不同配比给药,测定各组血液流变学:全血黏度(WBV);血沉(ESR)、红细胞聚集指数(HCT);凝血指标:血浆凝血酶原时间(PT)、部分活化凝血活酶时间(APTT)、血浆纤维蛋白酶含量(Fbg)、凝血酶时间(TT);(3)设置相应的桃仁-大黄药对(0∶1、1∶5、2∶5、2∶3、1∶1、3∶2、5∶2、5∶1、1∶0)9种不同配比给药,采用HPLC测定不同配比药对中没食子酸、(+)儿茶素、番泻苷B、蒽醌类中化学成分提取量的变化。结合桃仁-大黄药对的活血化瘀作用,以筛选其最佳配比。结果:(1)共整理桃仁-大黄药对活血化瘀方剂54首,桃仁-大黄配比在1∶7至3∶1间,但在1∶3至2∶1间出现的频率最高,达81.5%,其中1∶1出现的频次为17次,所有配比中最高,占31.5%;(2)桃仁-大黄药对(2∶3、1∶1、3∶2)均能改善大鼠的WBV、ESR、HCT、PT、APTT、TT、Fbg相关指标(P<0.05,P<0.01)。其中桃仁-大黄(1∶1)组对各个指标的改善更优;(3)与大黄对照组比较,1∶5、2∶5、2∶3桃仁-大黄不同配比样品中的没食子酸、(+)儿茶素、番泻苷B、蒽醌类提取量逐渐减少;1∶1样品中大黄的蒽醌类、鞣质类化学成分总提取量到达最低值。3∶2、5∶2、5∶1桃仁-大黄不同配比样品中的没食子酸、(+)儿茶素、番泻苷B、蒽醌类提取量升高,各类化学成分提取量(大黄酚-1-O-葡萄糖苷、大黄酸除外)高于大黄对照组。结论:古代文献数据显示桃仁-大黄药对活血化瘀最佳范围为2∶3至3∶2之间,配比1∶1较为集中。不同桃仁-大黄配比对寒凝血瘀大鼠模型的影响,其中2∶3、3∶2配比显著,1∶1配比更优。 Objective: To mine ancient literature on best ratio of Taoren-Dahuang to promoting blood circulation and removing blood stasis, and to analyze its effect on the rat model of cold congelation and blood stasis. Methods:(1)The prescription data of the ratio of ancient and modern application of Taoren-Dahuang for promoting blood circulation and removing blood stasis was mined.(2)The acute cold congelation and blood stasis rat model was replicated by subcutaneous injection of epinephrine with ice water bath method. Uniform design method was used to set 9 different dosing ratio of Taoren-Dahuang(0∶1, 1∶5, 2∶5, 2∶3, 1∶1, 3∶2, 5∶2, 5∶1, 1∶0). The blood rheology of each group was determined, such as the whole blood viscosity(WBV), erythrocyte sedimentation rate(ESR), erythrocyte aggregation index(HCT). The blood coagulation indexes were also determined, such as plasma prothrombin time(PT), activated partial thromboplastin time(APTT), plasma fibrinase content(Fbg), thrombin time(TT).(3)Different dosing ratio of the Taoren-Dahuang(0∶1, 1∶5, 2∶5, 2∶3, 1:1, 3∶2, 5∶2, 5∶1, 1∶0) were set, and HPLC was used to determine the contents of gallic acid,(+)catechin, sennoside B and anthraquinones extraction in chemical extraction. Combined with the effect of Taoren-Dahuang for promoting blood circulation and removing blood stasis, the optimal proportion was screened. Results:(1)A total of 54 prescriptions of Taoren-Dahuang for promoting blood circulation and removing blood stasis was concluded, excluding 5 ratios which appeared just once. The ratio of Taoren-Dahuang was between 1∶7 to 3∶1, but the highest frequency was between 1∶3 to 2∶1, which accounted for 81.5%. Among them, the frequency of occurrence of 1∶1 was 17, which was the highest and accounted for 31.5%.(2)Taoren-Dahuang for 2∶3, 1∶1 and 3∶2 could improve related indexes such as WBV, ESR, HCT, PT, APTT, TT, Fbg(P〈0.05, P〈0.01). Taoren-Dahuang 1∶1 group improved all indexes most obviously.(3)Compared with the Dahuang group, the contents of gallic acid,(+)catechin, sennoside B and anthraquinones extraction in 1∶5, 2∶5 and 2∶3 Taoren-Dahuang groups gradually reduced; anthraquinones and tannins extraction of Dahuang in 1∶1 Taoren-Dahuang group was the lowest. The gallic acid,(+)catechin, sennoside B and anthraquinones extraction in 3∶2, 5∶2 and 5∶1 Taoren-Dahuang groups increased, and the quantity of kinds of chemical extraction(rhubarb phenol-1-O-glucoside, except the rhein) was higher than that of Dahuang in the control group. Conclusion: Ancient literature data show that the best range of Taoren-Dahuang for a promoting blood circulation and removing blood stasis is between 2∶3 and 3∶2, and the ratio of 1∶1 is more concentrated. Different ratios of Taoren-Dahuang have effects on the cold congelation and blood stasis rat model, with 2∶3 and 3∶2 ratio being significant, and 1∶1 ratio being the most significant.
