poly(I:C)刺激鸡胚胎成纤维细胞产生外泌体的抗新城疫病毒活性 被引量:3

Activity of poly (I:C) in stimulating chicken embryonic fibroblasts to produce exosomes against Newcastle disease virus

摘要 外泌体(exosomes,Ex)是由各种细胞分泌的微小囊泡,可以通过传递RNA、DNA及蛋白质改变受体细胞状态。poly(I:C)是一种dsRNA模拟物,具有一定的抗病毒活性。为研究经poly(I:C)处理后鸡胚胎成纤维细胞(chicken embryo fibroblast,CEF)来源Ex的抗新城疫病毒(NDV)活性,通过超速离心法分离得到CEF来源Ex,并通过透射电镜和免疫印迹试验(Westernblot)对其进行鉴定,随后评估其抗NDV活性及活化靶细胞抗病毒能力。结果发现,poly(I:C)刺激使CEFEx分泌量增加1倍,且poly(I:C)Ex表现出比PBS Ex更强的抗NDV作用。进一步研究表明,poly(I:C)刺激CEF产生的Ex可通过TLR3-NFκB途径产生抗NDV作用。 Exosomes are microvesicles secreted from all types of ceils, can change the stage of recipient cells by transmitting intracellular information including DNA,RNA and protein, poly(I:C) is the simulator of dsRNA,and stimulates the anti-viral activity. In order to study the anti-Newcastle virus(NDV) activity of the exosomes of the chicken embryonic fibroblasts(CEF) after poly(I: C) treatment, the exosomes of the CEF source were isolated by speeding centrifugation and identified by transmission electron microscopy(TEM) and Western blot,the activity resist NDV and target cells were then assessed. The results showed that poly(I:C) stimulation doubled CEF exosome secretion,and the poly(I:C) Ex exhibited a more potent anti-NDV activity than PBS Ex. In further investigation,TLR3-NFκB pathway was involved in this process. In general, exosomes from poly (I:C) stimulated CEF performed a potent anti-NDV activity through TLR3-NFκB pathway.
