iTRAQ技术分析无乳链球菌牛源株与人源株差异表达蛋白 被引量:5

Analysis of different expression proteins in Streptococcus agalactiae between bovine and human strain by iTRAQ

摘要 为了分析无乳链球菌牛源株ATCC13813与人源株A909差异蛋白质组,提取ATCC13813与A909全菌蛋白,iTRAQ技术标记后,进行液相色谱串联质谱(LC-MS/MS)鉴定和定量,质谱数据通过Mascot2.2和Proteome discoverer 1.4软件分析,并对差异蛋白进行GO功能注释和KEGG pathway通路分析。结果显示,共鉴定出差异表达蛋白350个(P〈0.05),与ATCC13813相比,在A909中上调表达蛋白174个(比值〉1.5),下调表达蛋白176个(比值〈0.667)。生物信息学分析预测这些蛋白主要涵盖28个生物学功能,14个通路。本研究为阐明不同宿主来源株无乳链球菌致病性差畀奠定基础。 To identify different expression proteins in Streptococcus agalactiae between bovine (ATCC13813)and human strain(A909), cellular proteins were extracted from ATCC13813 and A909,and labelled with iTRAQ reagent. Different expression proteins were identified and quantified with LC-MS/MS. The mass spectrometry data was analyzed by Mascot 2.2 and Proteome discoverer 1. 4, and then subjected to biological information analysis of GO (gene ontology) and KEGG pathway. A total of 350 differentially expressed proteins were revealed(P〈0.05), among which,174 proteins were up-regulated(ratio〉1. 5) and 176 proteins were down-regulated(ratio〈 0. 667) in A909 compared to ATCC13813. Bioinformatics analysis predicted that these proteins were mainly involved in 28 biological functions and 14 pathways. Different expression proteins were identified in S. agalactiae bovine strain compared to human strain. These results may shed light on the difference of pathogenic mechanism of S. agalactiae strains isolated from different hosts.
