Protective Effects of Dandelion on Ethanol-induced Gastric Ulcer in Mice through Antioxidant Activity

摘要 目的:观察蒲公英对乙醇诱导的小鼠实验性胃溃疡的保护效果,并探讨其可能机制。方法:小鼠随机分为正常对照组、模型组和蒲公英高、中、低剂量组[15、5、1.5 g/(kg·d)],每组10只。各组灌胃给药1次/d,连续7 d。末次给药后,灌胃无水乙醇8 m L/kg诱导实验性胃溃疡模型。造模4 h后,分析蒲公英对模型小鼠胃溃疡指数、损伤抑制率,血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)和一氧化氮(NO)含量的影响。结果:与正常对照组比较,模型组小鼠胃黏膜溃疡指数显著上升,血清SOD活性下降,MDA和NO含量增加(P<0.01)。相对于模型组,蒲公英15、5 g/(kg·d)组小鼠胃黏膜损伤指数显著降低(P<0.01),血清SOD活性增强(P<0.01或P<0.05),MDA和NO水平降低(P<0.01)。结论:蒲公英对乙醇诱导的实验性胃溃疡具有良好的保护效应,其作用机制可能与增强机体抗氧化能力,抑制过量NO产生相关。 Objective: To evaluate the protective effect of dandelion on ethanol induced gastric ulcer and investigate its possible mechanism in mice. Methods: A total of 50 mice were randomly divided into normal group,model group,dandelion high,medium and low dose groups [15,5,1. 5 g/( kg·d) ],with 10 mice in each group. Administration of dandelion prior to ethanol ingestion dose-dependently inhibited gastric ulcer. After 4 h modeling,the gastric mucosal lesion was assessed by ulcer area,and mucosal injury index and injury inhibition rates were calculated. The contents of malondialdehyde( MDA) and nitric oxide( NO) in serum were measured. The activity of superoxide dismutase( SOD) was also determined. Results: Compared with the control group,the mucosal injury index,contents of MDA and NO in serum were markedly increased in the model group( P 〈 0. 01),and the activity of SOD was significantly decreased in mice model with gastric ulcer( P 〈 0. 01). Compared with the model group,dandelion [15,5 g/( kg·d) ] significantly reduced the gastric ulcer index,raised the activity of SOD in serum( P 〈 0. 01 or P 〈 0. 05). In addition,dandelion also significantly inhibited MDA and NO concentrations in serum in the mice exposed to ethanol induced ulceration( P 〈 0. 01 or P 〈 0. 05). Conclusion: Dandelion has good protective efficacy in treating ethanol induced gastric ulcer in mice. Its mechanism might be related to enhancing endogenous antioxidant capacity and inhibition of overproduction of NO.
