摘要
目的研究复方熊胆茵陈颗粒对非酒精性脂肪性肝炎(NASH)大鼠肝组织中TNF-α表达的影响。方法将60只SD大鼠随机分为空白组、模型组、西利宾安组、低剂量组、中剂量组和高剂量组,除空白组外予以高脂饲料喂养6周,每周1次后肢皮下注射40%CCL4花生油溶液制备脂肪性肝炎大鼠模型。空白组、模型组予以生理盐水灌胃,西利宾安组予西利宾安灌胃,低剂量组、中剂量组、高剂量组予复方熊胆茵陈颗粒按相应剂量灌胃。干预4周后取材,HE染色观察大鼠肝组织病理变化,PCR法和免疫组化法检测肝脏TNF-α的表达。结果复方熊胆茵陈颗粒能减少肝细胞的脂肪变性及炎细胞浸润,显著抑制TNF-α的表达(P<0.01)。结论复方熊胆茵陈颗粒可降低TNF-α的表达,这可能是其治疗NASH的作用机制之一。
Objective: To study the effect of Compound Xiongdan Yinchen Granules on expression of inflammatory eytokines TNF-α in rats with non-alcoholic steatohepatitis (NASH). Methods: Sixty SD rats were randomly divided into blank group, model group, western medicine group, low-dose group, middle-dose group and high-dose group. Except for the rats belong to blank group, every rat was fed to high fat feedstuffin 6 weeks and injected 40% CCL4 hypodermically once a week to establish NASH models. The blank group and model group were given saline by gavage, the westem medicine group were given silibin meglumine by gavage, and the low-dose group, middle-dose group and high-dose group were given compound Xiongdan Yinchen Granules by gavage. Aider 4 weeks, the rats were executed and liver were used for HE staining, while RT-PCR and immunohistochemistry were used to detect the relative expression levels of mRNA and protein of TNF-α. Results: Compound Xiongdan Yinchen granules can reduce hepatic steato- sis and inflammatory cell infiltration, and significantly inhibit the expression of TNF-α (P〈0.01). Conclusion: Compound Xiongdan Yinehen Granules can reduce the expression of TNF-α, which may be one of the mechanisms of its treatment for NASH.
