摘要
目的:探讨黄芪多糖对缺血再灌注损伤脐静脉内皮细胞氧化应激的影响。方法:将培养至5代的脐静脉内皮细胞随机分为正常对照组、模型组、模型+黄芪多糖组、模型+美托洛尔组,观察黄芪多糖对缺血再灌注损伤脐静脉内皮细胞活性、ROS、LDH、MDA含量、线粒体膜电位的影响。结果:与正常对照组比较,缺血再灌注损伤脐静脉内皮细胞ROS、MDA含量、LDH漏出率均显著升高,线粒体膜电位、细胞活性下降;黄芪多糖干预后,缺血再灌注损伤脐静脉内皮细胞ROS、MDA含量、LDH漏出率降低,线粒体膜电位、细胞活性升高。结论:黄芪多糖可能通过抑制自由基、脂质过氧化产物的生成,减少氧化应激而保护缺血再灌注损伤内皮细胞。
Objective: To observe the effect of astragalus polysaccharides on oxidative stress induced by human umbilical vein endothelial cells exposed to ischemia/reperfusion injury. Methods: Human umbilical vein endothelial cells cultured to 5 generation were divided into normal control group、model group、model + APS group、model + Metoprolol group.To observe the effect of APS on the cell viabilities、the content of ROS、MDA,the ratio of LDH leaked from cells,and mitochondrial membrane potential in HUVEC exposed to hypoxia/reoxygen. Results: The content of ROS、MDA and the ratio of LDH leaked from cells in the human umbilical vein endothelial cells exposed to ischemia-reperfusion injury were significantly higher than those in the normal control group,the mitochondrial membrane potential and the cell viabilities were lower than that in the normal control group; After giving the astragalus polysaccharides,the contents of ROS、MDA and ratio of LDH leaked from cells were significantly lower than those in model group,the cell viabilities and the mitochondrial membrane potential were higher than that in model group. Conclusion: Astragalus polysaccharides can protect human umbilical vein endothelial cells of ischemia-reperfusion injury,which may be related to that APS can reduce the production of free radicals and lipid peroxidation products in vascular endothelial cells and inhibit the oxidative stress.
