摘要
目的研究黄桑胶囊治疗慢性阻塞性肺疾病(COPD)模型大鼠的机制。方法将72只大鼠按体重随机均分为正常组、模型组、阳性组、黄桑胶囊(14.8、7.4、3.6 g·kg-1)组;麻醉大鼠后向其气管内注入0.2 m L脂多糖溶液,正常组注入0.2 m L生理盐水,每周一次,连续8周。于造模第29日起,ig给予阳性对照组大鼠固本咳喘片(0.77 g·kg-1),ig给予黄桑胶囊各剂量组相应浓度的黄桑胶囊,ig给予正常组、模型组大鼠生理盐水。于给药第42日采血,检测血清中白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)的水平。结果黄桑胶囊不同剂量组大鼠的一般情况明显好于模型组的;与模型组比较,黄桑胶囊高、中剂量组的IL-8水平显著下降,高剂量组的TNF-α水平也显著下降。结论黄桑胶囊对COPD的治疗作用与调节血清中IL-8、TNF-α的水平有关,可能是其干预COPD发生、发展的机制之一。
OBJECTIVE To study the mechanism of Huangsang capsule on the treatment of chronic obstructive pulmonary disease(COPD) model rat. METHODS 72 rats were randomly divided into normal group,model group,positive control group and Huangsang capsule(14. 8,7. 4,3. 6 g·kg(-1)) group. After anesthesia,rats models were prepared by intratracheal injection of 0. 2 m L lipopolysaccharide solution,while the rats of normal group were given 0. 2 m L saline,once a week for totally eight weeks. On the 29 th day after the model building,the positive group rats were gavaged with Guben Kechuan tablet(0. 77 g·kg(-1)),the rats of Huangsang capsule group were gavaged with Huangsang capsule of corresponding dose,while the rats of normal group and model group were given normal saline.On the 42 th day after gavaged,the levels of IL-8 and TNF-α in the serum were detected. RESULTS The general situation of rats in different dose of Huangsan capsule groups were better than that of model group. Compared with the model group,the levels of IL-8 in high and medium dose of Huangsan capsule groups were significantly decreased. And the levels of TNF-α were also significantly decreased in high dose of Huangsan capsule group. CONCLUSION The therapeutic effect of Huangsang capsule on COPD is related to the regulation of the level of IL-8 and TNF-α in the serum,which is one of the intervention of COPD occurrence and development mechanism.
