摘要
目的:通过响应面法对黑骨藤指纹图谱液相色谱方法进行研究,得到优化的色谱条件。方法:采用响应面法优化液相色谱条件的6个主要参数(流动相梯度、洗脱时间、流速、柱温、改性剂浓度以及进样体积)。结果:优化得到黑骨藤的色谱分析的条件为:Kromasil C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-水,梯度洗脱,时间50 min,流速1.5 m L/min,柱温50℃。结论:利用响应面分析法成功优化了黑骨藤指纹图谱的液相色谱条件。
Objective: To investigate the liquid chromatography fingerprint of Periploca forrestii by response surface method,to obtain the optimized chromatographic condition. Methods: Response surface optimization software Minitab17. 1. 0 were used to optimize six major parameters(mobile-phase gradient,elution time,flowrate,column temperature,modifier concentration and sample volume).Results: Optimization of chromatographic analysis of Periploca forrestii was as follows,the chromatographic column was Kromasil C18 column(250 mm×4. 6 mm,5 μm),which was based on a mobile phase of acetonitrile-water at the flow rate of 1. 5 m L/min. The detection time was 50 min and the column temperature was 50 ℃. Conclusion: Successful optimization of Periploca forrestii liquid chromatographic fingerprint is obtained by response surface methodology.
