摘要 目的 探讨 miR-21调控人唾液富组蛋白1 (HTN1)对肺癌细胞迁移和侵袭的影响。方法 定量聚合酶链反应和 Western blot分别检测肺癌组织和癌旁正常组织中 miR-21和 HTN1表达情况;使用双荧光素酶实验检测 miR-21和 HTN1之间的相互调控关系;划痕愈合实验检测 miR-21对肺癌细胞迁移能力的影响;Transwel侵袭实验检测 miR-21对肺癌细胞侵袭能力的影响。结果 与癌旁正常组织比较,肺癌组织中 HTN1蛋白的表达水平较高,miR-21mRNA的相对表达量较高;双荧光素酶检测miR-21和HTN1之间有直接的结合位点,miR-21可以调控HTN1的表达活性;划痕愈合实验显示抑制 miR-21的表达后 H1650细胞的迁移能力受到相应的抑制;Transwel侵袭实验显示抑制miR-21的表达后 H1650细胞的侵袭能力受到相应的抑制。结论 miR-21可以调控 HTN1蛋白的表达,影响 H1650肺癌细胞的迁移和侵袭行为。 Objective To investigate the effect of miR-21 on the migration and invasion of lung cancer cells. Methods The expressions of miR-21 and human saliva rich in tissue 1 (HTN1) in lung cancer tissues and adjacent normal tissues were detected by quantitative polymerase chain reaction and Western blot. The relationship between miR-21 and HTN1 was measured by double luciferase assay. The effect of miR-21 on the migration ability of lung cancer cells was examined by scratch healing. Transwell invasion assay was used to detect the effect of miR-21 on lung cancer cell invasion. Results Compared with adjacent normal tissues, the expressions of HTN1 protein and miR-21 mRNA in lung cancer tissues were higher, Double luciferase assay showed a direct binding site between miR-21 and HTN1. miR-21 could regulate the expression activity of HTN1. Scaling healing experiment showed that the migration of H1650 ceils was inhibited by the inhibition of miR-21 expression. Transwell invasion assay showed that the invasion of H1650 cells was inhibited by the inhibition of miR-21 expression. Conclusions miR-21 can regulate the expression of HTN1 protein and affect the migration and invasion of lung cancer ceils.
