己糖激酶1及NLRP3信号通路内关键分子在难治性根尖周炎中的表达研究 被引量:4

Expression of hexokinase 1 and key molecules of NLRP3 signaling pathway in refractory apical periodontitis

摘要 目的:检测难治性根尖周炎中己糖激酶1(hexokinase 1,HK1)及NLRP3信号通路内关键分子的表达情况。方法:选取20例需行根尖手术治疗的难治性根尖周炎患者,收集手术过程中刮除的肉芽组织;并收集20例正常根尖周组织。采用qRT-PCR法检测两组中HK1、NLRP3、Caspase-1、IL-lβ、IL-18表达情况。结果:难治性根尖周炎组中HK1、NLRP3、Caspase-1的mRNA水平明显高于正常组,而IL-lβ和IL-18 mRNA表达水平在两组间差异无统计学意义。结论:HK1/NLRP3/Caspase-1在难治性根尖周炎中可能起重要作用,但可能存在多个信号通路共同调控IL-1β和IL-18,在难治性根尖周炎中起免疫防御作用。 Objective: To detect the expression of hexokinase 1( HK1) and key molecules of NLRP3 signaling pathway in refractory apical periodontitis. Methods: Twenty patients with refractory apical periodontitis who underwent apical surgery were selected to collect the granulation tissue scraped during the operation. And twenty normal periapical tissues were collected from the ambulatory impacted teeth. The expressions of HK1,NLRP3,Caspase-1,IL-1β and IL-18 in the two groups were detected by qRT-PCR. Results: qRT-PCR showed that the mRNA levels of HK1,NLRP3 and Caspase-1 in refractory apical periodontitis were significantly higher than those in normal group,while there was no significant differences in mRNA levels of IL-1β and IL-18 between the two groups. Conclusion: HK1/NLRP3/Caspase-1 may play an important role in refractory apical periodontitis,but there may be multiple signaling pathways co-regulate IL-1β and IL-18,and play an immune defense effect in refractory apical periodontitis.
