滤纸药膜快速检测家蝇抗性方法的研究 被引量:3

摘要 目的研究家蝇对高效氯氰菊酯和氯氰菊酯抗药性的快速检测方法。方法采用滤纸药膜法,即将涂抹药液的滤纸放入烧杯中,并将试虫置于滤纸药膜内,观察试虫的死亡情况,并将检测结果与点滴法和玻璃瓶药膜法的结果进行对比分析。结果以滤纸药膜法分别筛选出高效氯氰菊酯LC99及氯氰菊酯LC95所对应的浓度作为诊断剂量,剂量值分别为45.49和30.28μg/cm2,两种药剂对应的诊断剂量下敏感品系家蝇的校正死亡率分别为98.89%和95.55%;两个野外品系家蝇(野外品系1和野外品系2)的校正死亡率分别为5.56%、44.45%和37.78%、7.78%;高效氯氰菊酯和氯氰菊酯滤纸药膜在4℃下保存1个月之久,仍可保持其稳定的检测灵敏性。结论本研究中的滤纸药膜法操作简单,2 h即可获得检测结果,为家蝇抗性的现场检测提供了一种快速方法。 Objective To study the rapid detection method for resistance of Musca domestica to beta-cypermethrin and cypermethrin. Methods The filter paper residual film method was applied. The filter paper evenly coated with the insecticide solution and put it into beaker. Musca domestica was placed in the filter paper residual film and the mortality was observed at the appropriate time. The results were compared with the results of topical application method and glass bottle residual film method. Results The concentration of beta-cypermethrin LC99 and cypermethrin LC95 were screened by the filter paper residual film method as the diagnostic dose,the dose values were 45. 49 and30. 28 g/cm2 respectively. The corrected mortality of the sensitive strains Musca domestica of the two insecticide were98. 89% and 95. 55%. The correction mortality of two wild strains Musca domestica( field strain 1 and field strain 2)were 5. 56%,44. 45%,37. 78% and 7. 78% respectively. Storage test showed that the filter paper residual film method of beta-cypermethrin and cypermethrin could be stored up to one month at 4 ℃. Conclusion The bioassay method used in this study has the advantages of simple operation and rapid detection which can provide a quick measurement method for the field detection of Musca domestica resistance.
