摘要
目的探讨高脂饲料联合链脲佐菌素(STZ)诱导建立稳定且长期维持的2型糖尿病小鼠模型的优化给药方案,确定最适STZ造模剂量。方法将20只C57BL/6雄性小鼠随机分为正常对照组、STZ 1组、STZ 2组和STZ 3组,每组5只。正常对照组给予普通饲料喂养,STZ 1、2、3组均给予高脂饲料喂养。喂养4周后,正常对照组腹腔注射0.1mL柠檬酸缓冲液,STZ 1、2、3组分别按照3、4、5mL·kg^(-1)腹腔注射1%STZ液,1次·d^(-1),连续3d。观察各组小鼠干预前和干预1~11周后的一般状况(摄食、饮水量、尿量及精神和毛发光泽)、体质量和血糖水平。结果与正常对照组比较,STZ 1、2、3组饮水量、进食量和排尿量增加,血糖均上升。造模11周结束时,STZ1组小鼠体质量显著高于正常对照组(P<0.01),且体质量增加值也高于正常对照组。STZ 2、3组小鼠STZ干预1~11周后体质量与正常对照组比较差异均无统计学意义(均P>0.05),但体质量增加值低于正常对照组;STZ 1、2、3组小鼠血糖值分别为(18.00±5.44)mmol·L^(-1)、(26.47±2.12)mmol·L^(-1)、(16.86±5.02)mmol·L^(-1),随机血糖均显著高于正常对照组(8.57±0.68)mmol·L^(-1)(P<0.05或P<0.01)。结论高脂饲料联合不同STZ剂量可诱导不同程度的糖尿病发生,诱导建立雄性C57BL/6小鼠长期稳定的2型糖尿病模型的最适1%STZ液剂量为4mL·kg^(-1)。
Objective To optimize the regimen of high-fat diet combined with streptozotocin(STZ)and to determine the optimum STZ dose for establishing a mouse model of long-term stable type 2 diabetes.Methods Twenty C57 BL/6 male mice were randomly divided into four groups,with 5 mice in each group.The normal control group was given general feed,while the STZ 1,STZ 2 and STZ 3 groups were treated with high-fat diet.After 4 weeks of feeding,normal control group was intraperitonealy injected with 0.1 mL citric acid buffer,and STZ 1,STZ 2 and STZ 3 groups were respectively given 1% STZ 3,4 and 5 mL·kg-1 once per day for 3 consecutive days.The general condition(food intake,water intake,urine output,spirit and hair luster),body weight and blood glucose level were observed before and after intervention for 1-11 weeks.Results Compared with normal control group,the water intake,food intake,urine output and blood glucose level increased in STZ-treated groups.At the end of 11 weeks of treatment,the body weight and body weight gain in STZ 1 group were significantly greater than those in normal control group(P〈0.01).There was no significant difference in body weight between normal control group and STZ 2 group,as well as between normal control group and STZ 3 group(P〉0.05).However,the body weight gain in STZ 2 group or STZ 3 group was lower than that in normal control group.The blood glucose level in STZ 1 group((18.00±5.44)mmol·L-1),STZ 2 group((26.47±2.12)mmol·L-1)or STZ 3 group((16.86±5.02)mmol·L-1)was obviously higher than that in normal control group((8.57±0.68)mmol·L-1)(P〈0.05 or P〈0.01).Conclusion High-fat diet combined with different doses of STZ can induce different degrees of diabetes.The optimal dosage of 1% STZ for inducing long-term stable type 2 diabetes is 4 mL·kg-1 in male C57 BL/6 mice.
