摘要
目的探讨Disabled-1(Dab1)在乳腺癌组织中的表达情况及其临床意义。方法采用免疫组织化学检测50例乳腺癌组织和相应癌旁正常组织中Dab1表达水平,并分析其表达与患者临床病理特征的关系;通过Oncomine数据库分析Dab1 m RNA在乳腺癌中的表达情况;下载GEO数据库中乳腺癌基因表达数据,采用Kaplan-Meier法分析Dab1的表达水平与乳腺癌患者预后的关系。结果乳腺癌组织中Dab1阳性表达率48.00%(24/50),癌旁组织中Dab1的阳性表达率为82.00%(42/50),两者阳性率的差异有统计学意义(χ~2=100.00,P=0.000);Dab1蛋白表达与乳腺癌患者的年龄、月经状态、肿瘤大小、病理类型无关(P>0.05),但与患者的肿瘤分化程度、TNM分期、淋巴结转移有关(P<0.05)。结论 Dab1蛋白在乳腺癌组织中表达降低,且与患者的肿瘤分化程度、TNM分期、淋巴结转移有关。
Objective To study the expression of Disabled-1(Dab1) protein in breast cancer and the potential correlation with clinical prognosis. Methods Totally 50 cases of patients with breast cancer were involved in this study. Expression of Dab1 in cancer tissue and nearby normal tissues was detected with immunohistochemistry. The relationship between expression of Dab1 and clinical characteristics was analyzed. m RNA levels of Dab1 was analyzed by tumor microarray database(Oncomine). Correlation between expression of Dab1 and prognosis of breast cancer was identified by Kaplan Meier curve. Results The positive rate of Dab1 in breast cancer in cancer group was significantly higher compared with normal group(48.00% vs 82.00%, χ^2 = 100.00, P = 0.000). Expression of Dab1 in breast cancer tissue was positively correlated with tumor differentiation degree, TNM staging and lymph node metastasis(P〈0.05). No obvious correlation was founded between Dab1 and age, menstruation, tumor size or pathological type of tumor. Conclusion Down-regulated of Dab1 breast cancer tissueis correlated with tumor differentiation degree, TNM staging and lymph node metastasis.
