摘要
目的 :观察加味理肠饮对腹泻型肠易激综合征(IBS-D)模型大鼠血浆、结肠组织5-羟色胺(5-HT)与神经肽(NPY)、丘脑及结肠组织5-羟色胺3受体(5-HT3R)mRNA与NPY mRNA表达、结肠肥大细胞(MC)水平的影响,以探讨其作用机制。方法:将SD大鼠随机分为7组,除空白对照组外,各组均予避水应激+夹尾刺激+番泻叶灌胃方法建立IBS-D模型,后分别给予相应药物灌胃,连续用药28 d后处死,运用Elisa法检测大鼠血浆及结肠组织中5-HT与NPY水平,并采用PCR法检测丘脑及结肠组织中5-HT3R mRNA、NPY mRNA的表达,采用甲苯胺蓝染色法检测MC的数量。结果:与空白组比较,模型组血浆及结肠组织5-HT的表达均上调、NPY表达下调(P〈0.05),模型组丘脑及结肠组织5-HT3R mRNA的表达均上调、NPY mRNA的表达下调(P〈0.05);与模型组比较,各药物组的血浆及结肠组织NPY表达均上调、丘脑及结肠组织5-HT3R mRNA表达均下调(P均〈0.05),除中药低剂量组外,其余各药物组的血浆及结肠组织5-HT表达均下调、丘脑及结肠组织NPY mRNA表达均上调(P〈0.05);与空白组比较,模型组结肠组织MC数增多(P〈0.05);与模型组比较,各药物组MC均减少(P〈0.05)。结论 :加味理肠饮能够有效的治疗IBS-D,其作用机制可能与调节5-HT、NPY、5HT3R mRNA、NPY mRNA的表达以及MC水平有关。
Objective:To observe the effects of Jiawei Lichang Decoction on 5-hydroxytryptamine(5-HT) and neuropeptide(NPY) in the plasma and colon tissues of rats with diarrhea-predominant irritable bowel syn- drome(IBS-D),5-HT3R mRNA and NPY mRNA expressions in thalamus and colon tissues and colon mast cell(MC) levels to investigate the mechanism. Metbods:SD rats were randomly divided into 7 groups. Ex- cept the blank control group,all groups were treated with avoiding water stress + tail-clip stimulation + senna gavage to establish the IBS-D model. Afterwards,the corresponding drugs were given by gavage re- spectively. After 28 days of treatment,the rats were sacrificed. The levels of 5-HT and NPY in rat plasma and colon tissues were detected by Elisa method. The expressions of 5-HT3R mRNA and NPY mRNA in thalamus and colon tissues were detected by PCR method and toluidine blue staining was used to examine the number of mast cells. Results:Compared with the blank group,the expression of 5-HT in the plasma and colon tissues in the model group increased and the NPY expression decreased (P〈0.05). The expressionof 5-HT3R mRNA in the thalamus and colon tis- sues increased and the expression of NPY mRNA decreased in the model group(P〈0.05). Compared with the model group,the expression of NPY in plasma and colon tissues increased and the expres- sion of 5-HT3R mRNA in the thalamus and colontissues decreased in all drug groups(P〈0.05). Except the low-dose herbal group,the expression of 5-HT in other groups decreased in both plasma and colon tissues,and the expression of NPY mRNA in the thalamus and colon increased(P〈0.05). Compared with the blank group,the number of mast cells in the colon of the model group increased(P〈0.05). Compared with the control group,the number of mast cells in all groups decreased(P〈0.05). Conclusion:Jiawei Lichang Decoction can effectively treat IBS-D,and its mecha- nism may be related to the regulation of 5-HT,NPY,5-HT3R mRNA,NPY mRNA expressions and mast cell levels.
