上海水体中抗生素抗性基因的分布特征 被引量:8

Distributive situation of the antibiotic resistance genes in the surface water bodies in Shanghai

摘要 为研究上海市水体中抗生素抗性基因(Antibiotic Resistance Genes,ARGs)的污染状况和分布特征,采集17个不同地段的水样,进行了耐药菌和ARGs丰度的检测。结果表明,水中磺胺嘧啶类耐药菌、氨苄青霉素类耐药菌、环丙沙星类耐药菌、氯霉素类耐药菌的耐药率分别为14.81%-94.55%、0-46.60%、0-91.07%、0-87.5%。养殖场周围河流总耐药菌量普遍高于非养殖场周围河流。非养殖场周围水体和养殖场周围水体中抗生素抗性基因sul1、amp R、qnr S、cml A均被检出。定量PCR的检测结果表明,非养殖场周围水体和养殖场周围水体中氯霉素类抗性基因含量最高,其次为磺胺类抗性基因,养殖场周围水体抗生素抗性基因含量普遍高于非养殖场周围水体。抗生素抗性基因sul1、amp R、qnr S、cml A的含量与微生物、温度、悬浮物等影响因素显著性相关(p〈0.05),表明在水环境中这些影响因子对抗生素抗性基因的分布影响较大。 This paper is aimed to present the study results of the contamination status-in-situi of the ARGs( Antibiotic Resistance Genes) at the surface water from Huangpu River,Shanghai. As a matter of fact,when the surface water samples of the River were collected,we have made an assay on the testing results of the contamination situation and the level of the ARGs with the resistant strain to be used as the basic data for the correlation analysis between the antibiotic resistance genes and the microorganisms,the p H value,SS,TOC,DO and the coresponding temperatures.Furthermore,we have also examined and determined the content concentration rates of the resistant strain( sulfonamide-resistant bacteria, ampicillin-resistant bacteria, ciprofloxacin-resistant bacteria and chloramphenicol-resistant bacteria) of the ARGs( sul1,amp R,qnr S and cml A) via the heterotrophic plate accounting( HPC) and Q-PCR in accordance with their respective levels of contamination so that it would be possible for us to identify and distinguish between the distribution characteristic features. At the same time,we have also chosen the ciprofloxacin and gene qnr S to trace the effects of the different concentrations of ciprofloxacin on the qnr S. Thus,the results of our investigation and analysis demonstrate that the resistance rates of the drug-resistant strain in the surface water have been made to account for14. 81%-94. 55%,0-46. 60%,0-91. 07%,0-87. 5% for the sulfonamide-resistant bacteria,ampicillin-resistant bacteria,ciprofloxacin-resistant bacteria and chloramphenicol-resistant bacteria,in respective correspondence. The content rates of the drug-resistant strain in the water can be described mainly in a descending order in the following manner: the rivers or brooks around the animal farms〉 those around the non-livestock pastures,and,those around the pig farms〉 cattle farms〉 chicken farms. Furthermore,we have also tested and determined that the content rates of sul1,amp R,qnr S and cml A genes in the rivers which have been found accounting for 10^4-10^10 copies/μL,10^3-10^9 copies/μL,10^2-10^7 copies/μL and 103-1010 copies/μL in the water,in respective correspondence. Besides,the antibiotic resistance gene content concentration rate in the water environment around the farm has been generally found higher than that around the non-livestock pastures. Of all,the sul1 genes in the aforementioned rivers and brooks turn out to be thickest as compared with the other kinds of genes. Thus,it can be concluded that the distribution features of the genes in the river water in the above mentioned areas turn to be similar to those of the drug-resistant strains. Thus,in summery,the distribution status in situ of the genes in the regions concerned tends to be roughly in a gradually decreasing trend from the east to the west of the River. The content rates of the resistance genes( sul1,amp R,qnr S and cml A) turn out to be significantly correlated with the following factors,such as the microorganism,temperature and suspension situation( p〈0. 05). All the above results can be used to clarify that the factors concerned may have a greater impact on the distribution of the resistance genes in the water environment. Hence,the given research can be expected to offer a great help to make clear the pollution situation of the drug-resistant strain and resistance genes in the surface water in Shanghai region and thus provide a valuable reference to further studies on the pollution of ARGs in Huangpu River,Shanghai.
