摘要
目的：探索适合果实类中药的通用DNA条形码鉴定方法。方法：以南五味子、五味子药材为实验材料,筛选最佳DNA提取方法和提取部位,选择ITS2片断进行PCR扩增后测序,所得序列用MEGA 5.10软件分析比对,计算遗传距离（K2P）,利用Neighbour-joining tree（NJ树）法和Blast法进行序列比对,构建系统聚类树;并用木瓜等8种常用果实类中药验证方法的可行性和有效性。结果：适合果实类中药的通用DNA条形码鉴定方法是：采用试剂盒法（离心柱型）提取种仁部位DNA,以ITS2片段为DNA条形码序列进行PCR扩增,琼脂糖凝胶电泳检测及测序后,录入＂中药材DNA条形码鉴定系统＂进行物种鉴定。结论：该方法可有效应用于果实类药材的鉴定。
Objective： To explore a general DNA barcode identification method for fruits Chinese drugs. Methods： Schisandra Sphenantherae Fructus and Schisandra Chinese Fructus samples were used to select the best DNA extraction method and extraction part. We selected ITS2 fragment for PCR. MEGA 5. 10 software was utilized for related data calculating. Then neighbour-joining tree（NJ tree） was built based on K2 P model and blast method was used to identify specie of samples in this study. And then 8 kinds of fruits medicine were used to verify the feasibility and effectiveness of the method. Results： The general DNA bar code identification method for fruits Chinese drugs was extracted DNA from kernel,selecting ITS2 fragment for PCR and searching species information in＂www. Tcmbarcode. cn＂after sequencing. Conclusion： The method can be effectively applied to the identification of fruits TCM.
