消癌解毒方对CT26荷瘤小鼠IL-6/STAT3信号通路的影响 被引量:16

Effect of Xiao’ai Jiedu Prescription on IL-6/STAT3 Signal Pathway in CT26 Tumor-bearing Mice

摘要 目的:观察消癌解毒方对荷瘤小鼠瘤组织内IL-6/STAT3信号通路的影响。方法:将40只雄性CT26荷瘤小鼠随机分为模型对照组(生理盐水灌胃2 m L·d^-1),消癌解毒方低、中、高剂量组(消癌解毒方中药灌胃5,10,20 g生药·kg^(-1)),奥沙利铂组(奥沙利铂腹腔注射5 mg·kg^-1),每组8只。连续给药14 d,末次给药2 h后处死小鼠,剥取瘤组织。采用末端转移酶标记技术(TUNEL)法观察小鼠瘤组织中细胞凋亡率;免疫组化染色法检测小鼠瘤组织中Cleaved Caspase-3蛋白表达;蛋白质免疫印迹(Western blot)法检测小鼠瘤组织中信号转导和转录激活因子3(signal transducers and activators of transcription 3,STAT3),磷酸化的信号转导和转录激活因子3(phospho-STAT3,p-STAT3),白细胞介素-6(interleukin-6,IL-6)蛋白的表达。结果:TUNEL结果显示,与模型对照组比较,消癌解毒方各组小鼠瘤组织中细胞凋亡指数明显升高(P〈0.01);免疫组化结果显示,与模型对照组比较,消癌解毒方中、高剂量组可以显著增加Cleaved Caspase-3蛋白表达(P〈0.05);Western blot结果表明与模型对照组比较,消癌解毒方中、高剂量组可以显著降低pSTAT3,IL-6的蛋白表达(P〈0.05)。结论:消癌解毒方可以促进肿瘤细胞凋亡,其机制可能与抑制IL-6/STAT3信号通路,上调Cleaved Caspase-3的表达有关。 Objective: To investigate the effect of Xiao'ai Jiedu Prescription on IL-6/STAT3 signal pathway in CT26 tumor-bearing mice. Methods: Forty male CT26 tumor-bearing mice were divided randomly into model control group( i. g. NS 2 m L·d^-1),Xiao'ai Jiedu Decoction low,medium and high dose groups( ig Xiao'ai Jiedu Prescription 5,10,15 g·kg^-1),oxaliplatin group( i. p. oxaliplatin 5 mg·kg^-1) with 8 mice in each group. Continuous administration was lasted for 14 days. After the final administration for 2 hours,mice were sacrificed and tumor tissues were stript. TUNEL assays was adopted to detect the apoptosisindex( AI) of tumor tissue. The proteins of Cleaved Caspase-3 in tumor tissue were detected by immunohistochemistry. Western blot was performed to detect the proteins of STAT3,p-STAT3 and IL-6 in tumor tissue. Results: Tunel results showed that compared with model control group,each Xiao'ai Jiedu Decoction group,s AI was high( P〈0. 01). The immunohistochemical staining results showed that the protein of Cleaved Caspase-3 were increased( P〈0. 05) in medium and high dose Xiao'ai Jiedu Decoction groups. And the Western blot results showed that Xiao'ai Jiedu Prescription decreased the expressions of p-STAT3 and IL-6 protein compared with the model control group( P〈0. 05). Conclusion: Xiao' ai Jiedu Prescription can improve the apoptosis in tumor tissue. The mechanism may be correlated with inhibition of IL-6/STAT3 signal pathway and up-regulation of the expression of Cleaved Caspase-3.
