归芍茶多酚方剂体内抗氧化作用实验研究

Study on Antioxidation of the Extract of Angelica and Peony with Tea Polyphenols

摘要 目的:评价归芍茶多酚方剂的抗氧化作用。方法:60只雄性小鼠分为对照组、模型组、阳性对照组及归芍茶多酚方剂3个剂量组,除对照组外D-半乳糖500 mg/kg腹腔注射造模,注射量为0.1 m L/10 g,每日1次,连续造模6周。给药组连续灌胃30 d,同时模型组和各给药组继续给予相同剂量D-半乳糖腹腔注射。末次灌胃1 h后,取血测定血清抗氧化能力。另取60只小鼠给药方法同前,末次灌胃后,禁食16 h,模型组和给药组灌胃给予50%乙醇12 m L/kg,6 h后取肝脏,测定肝组织的抗氧化能力。结果:归芍茶多酚方剂连续小鼠灌胃30 d,各剂量组血清SOD活力、GSH含量均显著升高(P〈0.01或P〈0.05),蛋白质羰基含量显著下降(P〈0.01),归芍茶多酚方剂高剂量组MDA含量显著下降(P〈0.05)。各剂量组肝组织SOD活力均显著升高(P〈0.01),MDA含量、蛋白质羰基含量显著下降(P〈0.01或P〈0.05),归芍茶多酚方剂高、中剂量组GSH含量显著上升(P〈0.01)。结论:归芍茶多酚方剂对动物具有抗氧化功效。 Objective:To evaluate the antioxidant effects of the extract of Angelica and Peony with tea polyphenols(TP). Methods:60 male mice were randomly divided into 6 groups:control group,model group,positive group and three dose groups of the extract of Angelica and Peony with TP. 500 mg/kg D-galactose were intraperitoneal injected in mice except for the control group,0.1 m L/(10 g·day),continuous 6 weeks. After ig. administration for 30 days,blood sample was collected and antioxidant capacity were tested. By the same administration for 30 days,another 60 mice were single oral administration with 50% ethanol 12 m L/kg,liver sample was taked and antioxidant capacity were tested after 6 hours. Results:After 6 weeks of treatment,the content of SOD and GSH in the serum of each dose group of the extract of Angelica and Peony with TP increased significantly(P〈0.01 or P〈0.05),and the content of protein carbonyl group decreased significantly(P〈0.01),and the content of MDA in the high dose group of the extract of Angelica and Peony with TP decreased significantly(P〈0.05). The SOD activity of liver tissue in each dose group of the extract of Angelica and Peony with TP increased significantly(P〈0.01). The content of MDA and protein carbonyl content decreased significantly(P〈0.01 or P〈0.05). The content of GSH in the high,middle dose group of the extract of Angelica and Peony with TP increased significantly(P〈0.01). Conclusion:the extract of Angelicaand Peony with TP can increase the activity of antioxidant.
