摘要
目的:研究以"肾脑相济"理论为指导的电针疗法对围绝经期抑郁症模型大鼠海马及其内环境的影响。方法:随机将40只雌性SD大鼠分成空白组、模型组、电针组和西药组,用去势和慢性不可预见性刺激相结合的方法建立围绝经期抑郁症大鼠模型,西药组予盐酸氯米帕明进行灌胃,电针组选取"百会""肾俞"和"三阴交"行电针刺激。观察比较各组大鼠糖水偏嗜和悬尾不动情况,ELISA法测定大鼠海马糖皮质激素受体(GR)、盐皮质激素受体(MR)的含量;尼氏染色法检测海马CA3区神经细胞数目及核仁数量、总面积、平均黑度、积分光密度。结果:与模型组相比,电针组和西药组糖水偏嗜率明显增加、悬尾不动时间显著缩短;GR和MR水平升高,CA3区神经细胞数目增多,胞内核仁数目、总面积、积分光密度、平均黑度均升高。结论:围绝经期抑郁状态下,海马内环境稳态被破坏,以"肾脑相济"理论为指导的电针疗法可以改善海马内环境,减少大鼠神经细胞的损伤,并为临床电针改善抑郁状态提供了有力依据。
Objective:To study the effect of electroacupuncture therapy on the hippocampus and its internal environment in the rat model of perimenopausal depression. Methods:40 SPF SD female rats were randomly divided into blank group,model group,EA group and clomipramine group. The rat models of perimenopausal depression were established by castration combined with chronic unpredictable stimulus. Medicine group was treated with clomipramine hydrochloride by intragastric administration,electro acupuncture group selected"Baihui"and"Shenshu"and"Sanyinjiao"for electroacupuncture. The behavior was measured by the sugar consumption experiments and the tail suspension test. ELISA method was used to determine the content of glucocorticoid receptor(GR)and mineralocorticoid receptor(MR)in hippocampus of rats. Nissl staining method was used to detect the number of nerve cells and nucleolus in hippocampal CA3 area. Results:Compared with model group,electroacupuncture group and Western medicine group,sucrose preference rate increased significantly,significantly shortened immobility time in tail suspension;increased GR and MR levels,CA3 nerve cell number increase,intracellular nucleolar number,total area,integral optical density and average density increased. Conclusion:In the perimenopausal depression state,the hippocampal homeostasis is destroyed. The electro acupuncture therapy guided by the theory of kidney and brain communication can improve the hippocampal environment and reduce the damage of nerve cells in rats,and provide a strong basis for clinical electroacupuncture to improve the state of depression.
