当归黄芪汤对糖尿病大鼠肾组织JAK2STAT3信号通路的影响 被引量:5

Effect of Danggui Huangqi Decoction on JAK2STAT3 Signaling Pathway in Renal Tissue of Diabetic Rats

摘要 目的探讨当归黄芪汤对糖尿病大鼠肾组织酪氨酸激酶-信号传导与转录激活因子(JAK2STAT3)信号通路的影响。方法选择清洁级Wistar大鼠90只,根据随机数字表法分为观察组、模型组和对照组,各30只。模型组及观察组大鼠建立糖尿病模型,对照组大鼠腹腔内注射相同剂量的柠檬酸液。建模成功后第2天,观察组给予当归黄芪汤,模型组及对照组给予等量的蒸馏水,均灌胃给药,干预12周。结果观察组及模型组大鼠的24h尿蛋白定量(24hPRO)、尿素氮(BUN)及血肌酐(SCr)均明显高于对照组,但观察组明显低于模型组(P<0.05);观察组及模型组大鼠肾组织的超氧化物歧化酶(SOD)水平均分别明显低于对照组,且观察组明显高于模型组(P<0.05);丙二醛(MDA)水平均明显高于对照组,且观察组明显低于模型组(P<0.05);观察组及模型组大鼠肾组织的磷酸化酪氨酸激酶(p-JAK2)及磷酸化信号传导与转录激活因子(p-STAT3)表达均明显高于对照组,且观察组明显低于模型组(P<0.05);经Spearman法分析相关性后显示,p-JAK2及p-STAT3均分别与24hPRO,BUN,SCr及MDA呈正相关(P<0.05),与SOD呈负相关(P<0.05)。结论当归黄芪汤可抑制糖尿病大鼠JAK2STAT3信号通路的表达,该作用与氧化应激损伤有关,可通过检测JAK2STAT3信号通路的激活情况评价糖尿病的治疗效果。 Objective To investigate the effect of Danggui Huangqi Decoction on tyrosine kinase-signal transducer and activator of transcription( JAK2STAT3) signaling pathway in renal tissue of diabetic rats. Methods Totally 90 clean Wistar rats were selected and divided into the observation group, the model group and the control group according to the random number table method, 30 rats in each group. The diabetic model was established for the model group and the observation group. The same dose of citric acid was injected into the abdominal cavity of the control group. 2d after the success of the modeling,the observation group was given Danggui Huangqi Decoction,the model group and the control group were given the same amount of distilled water,intragastric administration. All the rats were intervened for 12 weeks. Results The levels of 24h urine protein quantity, urea nitrogen( BUN) and serum creatinine( SCr) in the observation group and the model group were significantly higher than those in the control group,and those in the observation group were significantly lower than those in the model group( P〈0. 05). The superoxide dismutase( SOD) levels of the renal tissue in the observation group and the model group were significantly lower than that in the control group,but that in the observation group was significantly higher than that in the model group( P〈0. 05). The malondialdehyde( MDA) levels of the renal tissue in the observation group and the model group were significantly higher than that in the control group,but that in the observation group was significantly lower than that in the model group( P〈0. 05). The expression of phosphorylated tyrosine kinase( p-JAK2) and phosphorylated signal transducer and activator of transcription( p-STAT3) in the renal tissue of the rats in the observation group and the model group were significantly higher than that in the control group,but that in the observation group was significantly lower than that in the model group( P〈0. 05). After analysis of correlation by Spearman, p-JAK2 and p-STAT3 were respectively positively correlated with 24h urine protein quantity,BUN,SCr and MDA( P〈0. 05),but they were negatively correlated with SOD( P〈0. 05). Conclusion Danggui Huangqi Decoction can inhibit the expression of JAK2TAT3 signaling pathway in diabetic rats,and this effect is related to oxidative stress injury. The effect of the treatment of diabetes can be evaluated by detecting the activation of the JAK2STAT3 signaling pathway.
