摘要
为探究PI-like基因对野鸢尾花器官的花柱分枝和外轮花被片形成的作用,通过RACE方法,分离PI-like型花器官特征基因,并运用实时荧光定量PCR法检测其表达模式。结果表明:克隆得到野鸢尾2个PI-like基因,命名为Id PI1和Id PI2,其编码的氨基酸序列与MADS-box转录因子家族中的PI/GLO亚家族成员具有高度的同源性。Id PI1和Id PI2编码的氨基酸序列的同源性为80%。系统进化分析显示,Id PI1和IdPI2分布在进化树的两支,表明其功能可能发生了分化。相对定量检测结果显示,Id PI1和Id PI2的转录产物在花器官的各轮结构中均可检测到,但其表达量有差异。Id PI1在外轮花被片表达量相对较高,是内轮花被片的2.7倍,是花柱分枝的15倍;而Id PI2在外轮花被片、内轮花被片和花柱分枝的表达量差异不明显,外轮花被片是内轮花被片的1.4倍,是花柱分枝的2.7倍。2个Id PI基因对外轮花被片和内轮花被片的形成均有影响,且Id PI2对花柱分枝形成较Id PI1更为重要。
This study aimed to explore the molecular mechanism controlling flower development in Iris dichotoma Pall.,particularly morphology of style branches and petaloid outer tepals. Two PI-like MADS-box genes were isolated by RACE and their expression patterns were detected by qRT-PCR.The results showed that two PI-like genes were obtained by cloning,which were named IdPI1 and IdPI2. The amino acids sequence encoded by them were highly homologous to the members of PI/GLO-like MADS-box transcription factor family. The homology of IdPI1 and IdPI2 was 80%. The phylogenetic analysis showed that IdPI1 and IdPI2 were distributed in one branch separately in the tree,indicating that their function might be differentiated. The expression results showed that the transcripts of IdPI1 and IdPI2 were detected in each ring flower organ,but the expression levels were different. The expression of IdPI1 inoutertepals was relatively high,2. 7 times as much as that in the inner tepals,15 times as much as that in the style branches. The expression difference of IdPI2 in outer tepals,inner tepals and style branches was not obvious,and the expression quantity of outer tepals was 1. 4 times as much as that of the inner tepals and 2. 7 times as much as that of the and inner tepals,however,the effect of IdPI2 on the formation of style branches was greater than that of IdPI1.
