蜂胶小分子纳米水凝胶对家兔口腔溃疡的治疗作用 被引量:4

Therapeutic Effect of Propolis Small Molecule Nanometer Hydrogel on Oral Ulcer in Rabbits

摘要 采用溶胶-凝胶法制备一种蜂胶小分子纳米水凝胶,评价其对口腔溃疡家兔的治疗效果,观察其对口腔溃疡家兔血清白细胞介素-6(IL-6)和肿瘤坏死因子α(TNF-α)表达水平的影响,探明其作用机制。采用40%冰醋酸灼烧法制备家兔口腔溃疡模型,随机分为5组,空白组不给予治疗,蜂胶小分子纳米水凝胶低剂量组(GEL低剂量组)、中剂量组(GEL中剂量组)、高剂量组(GEL高剂量组)按1支/2次/d麻醉后给药,西瓜霜组按2 mg/2次/d麻醉后给药。在治疗过程中测量家兔口腔溃疡直径,观察空白组、GEL高剂量组、西瓜霜组在治疗前和治疗4 d、8 d后口腔黏膜病理组织变化,检测血清IL-6和TNF-α表达水平。结果表明:与空白组比较,GEL低剂量组(P<0.01)、GEL中剂量组(P<0.001)、GEL高剂量组(P<0.001)和西瓜霜组(P<0.001)家兔口腔溃疡直径明显缩小。GEL高剂量组平均愈合时间为(9.50±1.29)d,与西瓜霜组相比,差异不显著(P>0.05),溃疡部位组织病变明显减轻,治疗效果与西瓜霜组相当。免疫功能失衡是口腔溃疡最重要的发病机制,而IL-6和TNF-α与口腔溃疡免疫失衡有密切关系,GEL高剂量组与西瓜霜组都能使IL-6及TNF-α显著下降,因此其机制可能是通过降低血中IL-6及TNF-α等炎性细胞因子表达水平来促进溃疡的愈合。 A kind of propolis small molecular nanometer hydrogels was prepared by sol-gel method to evaluate the therapeutic effect of oral ulcer in rabbits,to observe the effect of oral administration on the expression level of serum interleukin-6( IL-6) and tumor necrosis factor-α( TNF-α) in rabbits with oral ulcer,and to study its action mechanism. Oral ulcer in rabbit model was prepared with40% acetic acid burn method,rabbits were randomly divided into 5 groups,blank group was not given any treatment,low dose group( GEL low-dose group),middle dose group( GEL middle-dose group) and high dose group( GEL high-dose group) of propolis small molecular nanometer hydrogel were treated twice a day after anesthesia,and watermelon frost group was treated twice a day and2 mg after anesthesia. During the treatments,the diameter of rabbit oral ulcer was measured,and the pathological changes of the oral mucosa were observed in the blank group,GEL high-dose group and watermelon frost group before and after treatment for 4 d and 8 d,and the levels of IL-6 and TNF-α were detected. Results show that compared with that of the blank group,oral ulcer diameter of GEL low-dose group( P〈0. 01),GEL middle-dose group( P〈0. 001),GEL high-dose group( P〈0. 001) and watermelon frost group( P〈0. 001) rabbits significantly decreased. The average healing time of GEL high-dose group was(9. 50±1. 29) d,compared with watermelon frost group,P〉0. 05,there was no significant difference. The ulcer site tissue lesion was significantly reduced,and the therapeutic effect was similar to that of watermelon frost. The imbalance of immune function is the most important pathogenesis of oral ulcer,while IL-6 and TNF-α are closely related to the immune imbalance of oral ulcer,GEL high-dose group and watermelon frost group can significantly reduce IL-6 and TNF-α. There may be such a mechanism as promoting the healing of ulcer by reducing the expression levels of inflammatory cytokines like IL-6 and TNF-α.
