摘要
目的探讨口腔扁平苔藓(OLP)患者口腔黏膜组织的细胞凋亡情况、凋亡蛋白B细胞淋巴瘤-2(Bcl-2)以及Bcl-2相关X蛋白(Bax)的表达。方法选择40例OLP患者作为观察组,并同期选择40例健康志愿者作为对照组,运用免疫组织化学法检测两组口腔黏膜组织的凋亡蛋白Bcl-2、Bax表达情况,同时运用脱氧核苷酸末端转移酶介导的d UTP缺口末端标记法检测上皮细胞的凋亡情况。结果 OLP患者口腔黏膜凋亡细胞增多,染色质边聚,且可见凋亡小体。观察组口腔黏膜Bcl-2、Bax表达水平及其阳性率、上皮细胞凋亡指数均高于对照组(均P<0.05)。结论 OLP患者口腔黏膜组织存在淋巴细胞凋亡异常,细胞凋亡可能与OLP的发病密切相关,口腔黏膜组织中Bcl-2及Bax的表达水平或可作为临床诊断OLP的重要参考指标。
Objective To explore the cell apoptosis and expressions of apoptotic proteins,B-cell lymphoma-2（ Bcl-2） and Bcl-2-associated X protein（ Bax）,in oral mucosal tissues of patients with oral lichen planus（ OLP）. Methods Forty patients with OLP were enrolled as observation group,and 40 healthy volunteers as control group. Immunohistochemical method was used to detect the expressions of apoptotic proteins,Bcl-2 and Bax,in oral mucosal tissues in the two groups,and terminal deoxynucleotidyl transferase-mediated d UTP nick-end labeling method to detect the apoptosis of epithelial cells. Results Among the OLP patients,increased apoptotic cells,chromatin margination,and apoptotic bodies were observed in oral mucosa. The expression levels of oral mucosa Bcl-2 and Bax,their positive rates,and apoptosis index of epithelial cells in the observation group were all higher than those in the control group（ all P〈0. 05）. Conclusion Abnormality of lymphocyte apoptosis exists in oral mucosal tissues of OLP patients. Cell apoptosis might be closely related to the pathogenesis of OLP. The expression levels of Bcl-2 and Bax in oral mucosal tissues might act as an important reference indicator for clinical diagnosis of OLP.
