α-酮戊二酸对慢性氨氮胁迫下草鱼肾Rh糖蛋白、热休克蛋白70和过氧化物酶增殖物激活受体γmRNA表达的影响

摘要 为探讨饲粮中添加α-酮戊二酸(α-KG)对慢性氨氮胁迫下草鱼(Ctenopharyngodonidellus)肾Rh糖蛋白、热休克蛋白70(HSP70)和过氧化物酶增殖物激活受体γ(PPARγ)mRNA表达的影响,选取初始体重为(24.79±0.11)g的草鱼270尾,随机分成3组:C组(对照组,养于曝气后自来水中并饲喂基础饲粮)、T1组(氨氮组,养于18.37 mg/L的氨氮水中并饲喂基础饲粮)和T2组(α-KG组,养于18.37 mg/L的氨氮水中并饲喂0.75%α-KG饲粮)。氨氮胁迫后1、7、42 d进行采样,测定血氨含量及肾Rhag、Rhcg1、Rhcg2、HSP70和PPARγ的mRNA表达量。结果表明:氨氮胁迫下第1天,T1组血氨含量较对照组显著升高了21%(P<0.05),而日粮α-KG的添加能够有效缓解血氨含量的升高。肾Rhag、Rhcg1和HSP70 mRNA表达量在氨氮胁迫下出现显著差异。其中,氨氮胁迫后1 d,T1组Rhag mRNA表达量较对照组升高了348%(P<0.05),而α-KG的添加能够缓解该现象。氨氮胁迫7 d后,T1组Rhcg1 mRNA表达量与对照组无显著差异,T2组Rhcg1 mRNA的表达量较T1组显著降低了217%(P<0.05)。氨氮胁迫42 d后,T1组和T2组Rhag和Rhcg1 mRNA的表达量均较对照组显著升高(增量分别为405%和833%、59%和48%)(P<0.05)。此外,与T1组相比,T2组肾中HSP70的mRNA表达量显著降低了603%(P<0.05)。而氨氮胁迫7 d后,T1组和T2组HSP70 mRNA表达量较对照组显著降低,且减量分别为408%和404%(P<0.05)。综上所述,饲粮α-KG的添加能够有效缓解前期草鱼血氨含量的升高,并调节氨氮胁迫下Rhag、Rhcg1和HSP70的mRNA表达量,有利于草鱼应对慢性氨氮胁迫。 The objective of this study was to determine the effects of dietary α-ketoglutarate(α-KG) on rhesus glycoproteins, heat shock protein 70(HSP70) and peroxisome proliferator aetivated receptor γ(PPARγ)mRNA expressions of grass carp(Ctenopharyngodonidellus) under chronic ammonia exposure.270 grass carps[initial weight,(24.79 ± 0.11)g] were randomly assigned to three groups:C group,exposed to tap water and fed basic diet;T1 group,exposed to 18.37 mg/L ammonia water and fed basic diet;T2 group,exposed to 18.37 mg/L ammonia water and fed 0.75% α-KG added diet. Grass carps were sampled on day 1,7 and 42,and then the plasma ammonia content and kidney Rhag,Rhcg1,Rhcg2,HSP70,PPARγ mRNA expressions were measured.Results showed that the ammonia level in the T1 group was significantly higher than that in the control group by21% on day 1(P〈0.05),and dietary α-KG alleviated plasma ammonia increased.The kidney Rhag,Rhcg1 and HSP70 mRNA expression showed a significance difference among groups under ammonia exposure.On day 1,the Rhag mRNA expression of T1 group was significant increased 348%(P〈0.05) under ammonia exposure,and α-KG supplement could alleviate this phenomenon. On day 7,the Rhcg1 mRNA expression of C and T1 groups were no significant difference,and the Rhcg1 mRNA expression of T2 group was lower than that in the T1 group by 217%(P〈0.05).On day 42,the mRNA expression of Rhag and Rhcg1 were significant increased under ammonia exposure(the increments were 405% and 833%,59%and 48% respectively)(P〈0.05).Besides,the expression of HSP70 mRNA in T2 group was decreased than that in T1 group by 603%(P〈0.05).On day 7,The expression of HSP70 mRNA in T1 group and T2 group were significantly lower than that in the control group,and the reduction were 408% and 404%,respectively(P〈0.05).In conclusion,dietary α-KG could alleviate the increase of plasma ammonia content and adjust the mRNA expression of Rhag,Rhcg1 and HSP70 under ammonia exposure,which may be beneficial for grass carp to adapt choric ammonia exposure.
