摘要
以[C_nPy]Cl(n=2、4、6)-K_2HPO_4不同碳链长度的氯代吡啶类离子液体双水相为研究体系,利用清-浊点辅助法测定该体系的双节线及液液相平衡数据。采用Merchuk方程关联双节线,Othmer-Tobias、Bancroft方程关联液液相平衡数据。通过相关系数考察该体系上下相的氯代吡啶类离子液体浓度、上下相的K_2HPO_4浓度与蛋白浓度分配系数的相关度。结果表明:3个经验方程的拟合度分别在0.996、0.997、0.995以上,关联结果理想。木瓜蛋白酶分配系数的对数与上下相各组分浓度差的相关度均较高,故用Matlab建立其在该双水相中的分配模型,相对偏差都低于4%。相比其他同类模型,试验中建立的模型精确度提高,且能准确预测双水相中酶的分配行为。研究结果也可补充木瓜蛋白酶在双水相系统中的分配模型数据库。
The chlorinated pyridine ionic liquids aqueous two-phase with different carbon chain length [CnPy]Cl(n=2, 4, 6) -K2HPO4 were the investigative system. The binodal curve and liquid-liquid equilibrium weremeasured by means of clear-cloud points aid. The data fitting of binodal and the equilibrium were based onMerchuk,Othmer-Tobias and Bancroft equations. The relevant coefficient was used to investigate the proteincorrelation partition coefficient of papain with the concentration of the chlorinated pyridine ionic liquids in topand lower phase and theK2HPO4 concentration in top and lower phase. Results showed that the fitting degree ofthe three empirical equations was 0.996,0.997 and 0.995,respectively,which was ideal. Based on thelogarithm of the partition coefficient and the concentration difference between the two-phase components havinga high correlation,the partition model of papain in this [CnPy]Cl(n=2, 4, 6) -K2HPO4 system was established byMatlab. The relative deviation of this partition model was below 4%,which was less than other congenericmodel. The accuracy of this partition model was improved,which was sufficient to predict the partition behaviorof papain as well as supplemented partition models database in the aqueous two-phase system.
