摘要
目的探讨环境胁迫产生的活性氧能够增强植物的次生代谢改变药材的质量的方法。方法分别采用0.004μmol/L、0.4μmol/L和40μmol/L Na_2S_2O_4处理五味子鲜果,研究活性氧、抗氧化酶活性及木脂素类成分三者之间关系。结果 0.4μmol/L Na_2S_2O_4显著降低超氧化物歧化酶(SOD)活性,40μmol/L Na_2S_2O_4显著降低SOD活性;过氧化氢酶(CAT)活性变化主要发生在第1~2天和5~6天,与浓度呈负相关,0.004μmol/L和0.4μmol/L Na_2S_2O_4处理组增加CAT活性,40μmol/L处理组CAT活性变化很小;过氧化物酶(POD)活性变化主要发生在5~6天,三个浓度均显著增加;抗坏血酸氧化酶(APX)活性变化不明显。苯丙氨酸解氨酶(PAL)活性第5天和第6天显著升高,促进木脂素类成分生物合成,其中40μmol/L Na_2S_2O_4处理5 d最为显著,五味子醇甲、五味子醇乙、五味子甲素、五味子乙素、五味子丙素比传统加工方法分别提高24.6%、21.9%、28.0%、19.3%、27.3%。结论 Na_2S_2O_4改变了五味子抗氧化酶活性,提高了次生代谢水平,提高了质量。
Objective To explore increase of medicinal materials quality based on the reactive oxygen species from environmental stress induce the secondary metabolism. Methods The fresh fruits are soaked with 0.004 μmol/L, 0.4 μmol/L, and 40 μmol/L Na2S2O4, respectively, the relationship between the H2O2, antioxidase system, and lignanoids is investigated. Results The activities of the superoxide dismutase(SOD) are increased for the 0.4 μmol/L and decreased for the 40 μmol/L. The change of catalase(CAT), appear at the 1 ~ 2 d and 5 ~ 6 d, had a negative correlation with the concentration, the CAT are increased for 0.004 μmol/L and 0.4 μmol/L, changed little for 40 μmol/L. The peroxidase(POD) are increased for 3 concentration at 5 ~ 6 d. The ascorbic oxidase(APX) changed little. Similar to the CAT and POD, the phenylalnine ammonialyase(PAL) are remarkably increased. The increased PAL enhanced the biosynthesis of lignanoids, the contents of schisandrin, schisandrol B, deoxyschizandrin, schizandrin B, and schisandrin C increased by 24.6 %, 21.9 %, 28.0 %, 19.3 % and 27.3 %, respectively. Conclusion Na2S2O4 changed antioxidase activities, increased the biosynthesis of lignanoids, and improved the quality of Fructus Schisandrae chinensis.
