旋毛虫丝氨酸蛋白酶抑制剂对旋毛虫体外入侵肠上皮细胞的影响 被引量:1

Effects of Trichinella spiralis serine protease inhibitors on the invasion of intestinal epithelial cells in vitro

摘要 为探究2种旋毛虫丝氨酸蛋白酶抑制剂Ts Ad SPI和Ts Ka SPI对旋毛虫体外入侵肠上皮细胞的影响,建立了旋毛虫-Caco-2体外侵染模型,采用IPTG诱导大肠杆菌表达这2种旋毛虫丝氨酸蛋白酶抑制剂蛋白并大量纯化,探究不同剂量、不同作用时间Ts Ad SPI和Ts Ka SPI单独及共同作用于侵染模型对旋毛虫体外入侵肠上皮细胞的影响。结果表明:Ts Ad SPI和Ts Ka SPI对旋毛虫入侵肠上皮细胞均有促进作用:2种蛋白单独作用时,剂量为100 g且作用时间为5 h时,入侵率最高;2种蛋白共同作用时,剂量为100 g且作用时间为3 h时,入侵率最高;并且这2种蛋白的单独作用效果优于共同作用效果,这2种蛋白共同作用产生拮抗效果。为进一步研究旋毛虫丝氨酸蛋白酶抑制剂对旋毛虫入侵宿主的影响奠定了基础。 To explore the influence of two Trichinella spiralis serine protease inhibitors Ts Ad SPI and Ts Ka SPI on T. spiralis invasion the intestinal epithelial cells in vitro,T. spiralis-Caco-2 in vitro infection model was established.To determine the effects of different joint dose and time of Ts Ad SPI and Ts Ka SPI on the T.spiralis invasion the intestinal epithelial cells of host in vitro when they added separately or together,Ts Ad SPI and Ts Ka SPI were induced and purified by Escherichia coli expression system.The results showed that both Ts Ad SPI and Ts Ka SPI promoted the invasion of T. spiralis into intestinal epithelial cells.The invasion rates ware the highest when the two proteins were separately added at 100 g for 5 h.And when the two proteins were added together at the dosage of 100 g for 3 h,the invasion rate was the highest;the effect of two proteins added separately was better when they added separately,the two proteins had antagonistic effects on the promotion of T. spiralis invasion.This study laid the foundation for the further study on the effects of two serine protease inhibitors on the invasion of T. spiralis in hosts.
