摘要
建立了测定茶叶中前花青素的高效液相色谱分析方法。样品经过甲醇提取,采用C18柱(4.6 mm×250 mm)进行分离检测,外标法定量,流动相为水/甲醇/异丙醇/甲酸(73/13/6/8),流速为1.0 m L/min,检测波长为525 nm。前花青素在25.0~400μg/m L范围内呈良好线性,线性相关系数为0.9998,回收率为88.7%~95.4%,相对标准偏差(RSD)为6.7%,检出限为0.3 mg/100 g。实验结果表明,该方法准确可靠,灵敏度较高和重现性较好,适用于茶叶中前花青素的定性和定量检测。
An analytical method for the determination of proanthocyanidins in tea was established by high performance liquid chromatograph. The samples were extracted with methanol and separated on a C18 column( 4. 6 mm ×250 mm) HPLC separation of proanthocyanidins was quantified by the external standard method. The mobile phase was water/methanol/isopropanol/formic acid( 73/13/6/8) at a flow rate of 1. 0 m L/min. Detection wavelength was 525 nm.The proanthocyanidins had a good linearity in the range of 25. 0 ~ 400 μg/m L with a correlation coefficient of 0. 9998.The recoveries of samples were in the range of 88. 7% ~ 95. 4% and the relative standard deviations( RSD) were 6. 7%.The detection limit was 0. 3 mg/100 g. Experimental results showed that the method had accuracy reliability and high sensitivity. It was suitable for the qualitative and quantitative detection of tea leaves.
