摘要
目的探讨盐酸多奈哌齐对血管性痴呆(vascular dementia,VD)大鼠海马CA1区神经细胞的保护作用及其可能作用机制。方法通过永久性结扎大鼠双侧颈总动脉的方法(2-VO)制作大鼠VD模型,随机将60只SD大鼠分为假手术组、模型对照组、药物治疗组,每组20只,各组再随机分为1月组、2月组。应用Morris水迷宫实验检测大鼠的学习记忆能力;采用免疫组织化学染色检测大鼠海马CA1区血管内皮生长因子(vascular endothelial growth factor,VEGF)及N-甲基D-天冬氨酸受体-2B(NMDAR-2B)的表达;用TUNEL染色检测神经细胞凋亡。结果与模型对照组比较,治疗组大鼠学习记忆力明显改善;治疗组大鼠海马CA1区NMDAR-2B及VEGF表达明显高于模型对照组;治疗组大鼠海马区神经凋亡细胞较模型对照组明显减少(P均<0.05)。结论盐酸多奈哌齐能改善VD大鼠的记忆力,并通过上调VD大鼠海马CA1区NMDAR-2B及VEGF的表达保护神经细胞。
Objective To explore the protective effect and its possible mechanism of donepezil hydrochloride on neural cells in hippocampal CA1 area of vascular dementia(VD)rats.Methods Sixty SD rats were randomly devided into three groups:sham operation group,VD model group and Donepezil treated group,with20 cases in each.VD model were established by occlusion of the bilateral common carotid arteries.10 rats of each group were sacrificed at 1 month and 2 months post operation.The water maze test were performed to examine the ability of learning and memory of the rats,the expression of NMDAR-2 B and VEGF in hippocampal CA1 were measured by immunohistochemical staining and neurons apoptosis was observed with terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling(TUNEL)staining.ResultsCompared with VD model group,the donepezil treated group of learning and memory ability increased significantly,and the expression of NMDA receptor 2 B and VEGF increased significantly in hippocampus CA1 area(P〈0.05),and the apoptotic cells in the hippocampus of the treatment group were significantly reduced(P〈0.05).Conclusion Donepezil hydrochloride can improve the memory of VD rats,and protect neurons by increasing the expression of NMDAR-2 B and VEGF in the hippocampal CA1 area of VD rats.
