摘要
目的:探讨自噬是否可以提高乳腺癌MCF-7细胞对紫杉醇的敏感性。方法:取MCF-7细胞,分为空白对照组、紫杉醇组、3-MA(自噬抑制剂)组和3-MA+紫杉醇组,采用流式细胞术检测细胞凋亡,Western blot检测自噬相关蛋白LC3B-Ⅰ、LC3B-Ⅱ和p62蛋白的表达情况。另取MCF-7细胞,用3-MA预处理细胞后,加入紫杉醇,采用CCK-8检测细胞增殖,以未行3-MA预处理的细胞为对照。结果:紫杉醇和3-MA均可促进MCF-7细胞凋亡,同时降低LC3B-Ⅱ/LC3B-Ⅰ的比值,升高p62蛋白的表达,紫杉醇和3-MA联用有协同作用(P<0.05)。加用3-MA后,紫杉醇对MCF-7细胞的增殖抑制作用增强(P<0.05)。结论:抑制自噬可增强MCF-7细胞对紫杉醇的敏感性。
Aim: To investigate whether autophagy could enhance MCF-7 cells' sensitivity to taxol. Methods: MCF-7 cells were divided into four groups,namely blank control group,taxol group,3-MA group,and 3-MA + taxol group,flow cytometry was used to detect cell apoptosis,and western blot was used to detect the expression of autophagy-related proteins LC3 B-Ⅰ,LC3 B-Ⅱ and p62. In another experiment,after MCF-7 cells were pretreated with 3-MA,taxol was added,cell proliferation was detected by CCK-8 assay,and the cells not pretreated with 3-MA were used as control. Results: Taxol and 3-MA both promoted the apoptosis of MCF-7 cells,reduced the ratio of LC3 B-Ⅱ/LC3 B-Ⅰ,and increased the expression of p62 protein. The combination of taxol and 3-MA had a synergistic effect( P〈0. 05). In addition,after adding 3-MA,the inhibitory effect of taxol on cell proliferation was enhanced( P〈0. 05). Conclusion: Inhibition of autophagy can enhance the sensitivity of MCF-7 cells to taxol.
