摘要
以狗牙根品种C299为材料,采用RACE方法从干旱处理的C299中获得1个脱水素基因DHNS。其核酸序列长度为495bp,编码164个氨基酸;氨基酸序列具有明显的脱水素类蛋白特征,由2个K片段、1个S片段和1个Y片段组成,属于YSK2型脱水素;蛋白结构分析结果显示,其为典型的无序蛋白。在E.coli和拟南芥中超表达脱水素基因DHNS,干旱、盐胁迫、高渗胁迫和酸碱的环境下转基因E.coli和拟南芥的生长速度均显著高于对照,宿主菌和拟南芥的抗逆能力显著增强。
In the study,Bermudagrass cv.C299 were taken as the test material,a dehydrin gene was got from C299 stressed by drought using RACE techniques.The length of the gene was 495 bp,encoding 164 amino acids.The amino acid sequence had distinct characteristics of dehydrins.It consisted of 2 Kfragments,1 Sfragments and 1 Yfragments,belonged to dehydrin of YSK2 type.The results of Circular Dichrosim showed that DHNS was a typical disordered protein.In addition,over expression of‘C299'dehydrin genes in E.coli and Arabidopsis enhanced tolerance in drought,salt stress,hypertonic stress and acid-base environment.The experimental results showed that dehydrin gene DHNShad strong and broadspectrum stress-resistant physiological function.
