摘要
目的探讨井穴刺血对急性脑缺血(ACI)大鼠神经功能、血清血小板衍生化生长因子(PDGF)、神经元特异化烯醇化酶(NSE)水平的影响。方法选择清洁级中老年Wistar大鼠150只,随机选取28只作为假手术组、其余122只大鼠采用经颞途径及其改良大脑中动脉阻断法建立ACI模型大鼠,选取建模成功的84只大鼠随机分为治疗组(井穴刺血处理)、空白对照组(不作处理)和西药组(腹腔注射尼莫地平处理)各28只,对比各组大鼠建模后3 h、24 h、72 h、7 d的神经功能评分、血清PDGF、NSE水平。结果造模后3h~造模后7d,空白对照组、西药组及治疗组的神经缺损功能评分显著的高于假手术组(P<0.05);造模后24h~造模后7d,西药组和治疗组大鼠的神经缺损功能评分均显著的低于空白对照组(P<0.05),西药组大鼠的神经缺损功能评分均显著的低于治疗组(P<0.05);造模后3h~造模后7d,空白对照组、西药组及治疗组的血清NSE、PDGF显著的高于假手术组(P<0.05);造模后3h~造模后7d,西药组和治疗组大鼠的血清NSE、PDGF均显著的低于空白对照组(P<0.05),西药组大鼠的血清NSE、PDGF均显著的低于和治疗组(P<0.05)。结论井穴刺血对ACI大鼠具有一定的神经保护作用,可有效降低血清中NSE及PDGF水平。
Objective To investigate the effects of hole puncture on nerve function and the expression of Serum Platelet derived growth factor (PDGF) and neuron specific enolase (NSE) in rats with acute cerebral ischemia (ACI). Methods A total of 150 mid- dle age and old age Wistar rats were selected. 28 rats were randomly selected as the sham operation group, and the other 122 rats were used to establish the ACI model rats by the temporal pathway and the improved middle cerebral artery occlusion. The 84 rats were ran- domly divided into the treatment group (well hole pricking treatment), the blank control group (no treatment) and Western medicine group. Group 28 ( intraperitoneal injection of nimodipine) , each group had 3 rats in each group. The scores of neurological function, serum PDGF and NSE were compared at 3 h, 24 h, 72 h and 7 d after modeling. Results From 3h to 7d after modeling, the scores of neurological deficits in the control group, western medicine group, and treatment group were significantly higher than those in the sham operation group ( P 〈0. 05 ) ; from 24 hours after modeling to 7 days after modeling, The neurological deficit scores of the western medicine group and the treatment group rats were significantly lower than those in the blank control group ( P 〈 0.05 ) , and the nerve defect function scores in the western medicine group rats were significantly lower than those in the treatment group ( P 〈 0.05 ) ; 7 days after model establishment, the expression of serum NSE and PDGF in the blank control group, western medicine group, and treatment group were significantly higher than those in the sham operation group ( P 〈 0.05 ) ; 3 hours after model establishment and 7 days after model establishment, the level of Serum NSE and PDGF of western medicine group and treatment group rats were significantly lower than those in the control group ( P 〈 0.05 ) , and serum NSE and PDGF in the western medicine group were significantly lower than those in the treatment group ( P 〈 0.05 ). Conclusion The pricking of acupuncture points has a certain therapeutic effect on ACI rats, which can effectively reduce the level of NSE and PDGF in serum, thereby achieving the purpose of promoting nerve function re- covery.
