摘要
目的探讨赖氨酰氧化酶(LOX)及其相互作用蛋白X射线修复交叉互补蛋白5(XRCC5)在乳腺癌发生、发展中的作用。方法选择乳腺浸润性导管癌患者170例,取手术切除的乳腺癌组织及其相应癌旁正常组织,采用免疫组化SP法检测LOX、XRCC5表达,分析LOX阳性表达与XRCC5阳性表达的关系及二者与患者临床病理参数和预后的关系。结果乳腺癌组织LOX、XRCC5阳性表达率及其相对表达量均高于癌旁正常组织(P均<0.05)。Spearman非参数相关分析显示,乳腺癌组织LOX阳性表达与XRCC5阳性表达呈正相关关系(r=0.536,P<0.01)。LOX相对表达量与乳腺癌腋窝淋巴结转移、临床分期有关(P均<0.05),与患者年龄、肿瘤直径、组织病理分级无关(P均>0.05)。XRCC5相对表达量与临床分期有关(P均<0.05),与患者年龄、肿瘤直径、腋窝淋巴结转移、组织病理分级无关(P均<0.05)。LOX阳性表达者和(或)XRCC5阳性表达者总生存率均低于LOX阴性表达者和(或)XRCC5阴性表达者(P均<0.05)。结论乳腺癌组织LOX、XRCC5表达升高,二者表达变化可协同促进乳腺癌侵袭和转移,并有可能作为乳腺癌预后评估的指标。
Objective To investigate the role of lysyl oxidase( LOX) and its interacting protein X-ray repair crosscomplementing protein 5( XRCC5) in the occurrence and development of breast cancer. Methods Breast cancer tissues and the corresponding adjacent normal tissues of 170 patients with breast infiltrating ductal carcinoma were collected. The expression of LOX and XRCC5 protein was detected by immunohistochemistry SP method. The relationship between LOX positive expression and XRCC5 positive expression and their relationships with clinicopathological parameters and prognosis were analyzed. Results The positive expression rate and relative expression of LOX and XRCC5 in the breast cancer tissues were higher than those in the adjacent normal tissues( all P〈0. 05). Spearman rank correlation analysis showed that LOX positive expression in the breast cancer tissues was positively correlated with XRCC5 positive expression( r = 0. 536,P〈0. 01). The relative expression of LOX was related to axillary lymph node metastasis and clinical stages of breast cancer( all P〈0. 05),but was not related to age,tumor diameter or histopathological grade( all P〈0. 05). The relative expression of XRCC5 was related to clinical stage( P〈0. 05),but was not associated with age,tumor diameter,axillary lymph node metastasis or histopathological grade( all P〈0. 05). The overall survival rate of patients with LOX positive expression and/or XRCC5 positive expression was lower than that of patients with LOX negative expression and/or XRCC5 negative expression( P〈0. 05). Conclusion The expression of LOX and XRCC5 in breast cancer tissues increases,the expression changes of them can synergistically promote the invasion and metastasis of breast cancer,and they may be used as indicators for prognosis evaluation of breast cancer.
