摘要
目的:建立HPLC同时测定丹灯通脑胶囊中葛根素、野黄芩苷、丹酚酸B与丹参酮ⅡA的方法。方法:采用HPLC法,色谱柱为Inerstil-ODS-SP(250 mm×4.6 mm,5μm);流动相为乙腈-0.1%磷酸水溶液梯度洗脱;流速1 m L·min^(-1);检测波长为280 nm;柱温为30℃。结果:葛根素、野黄芩苷、丹酚酸B、丹参酮ⅡA分别在3.85~115.41μg·m L^(-1)(r2=0.999 8)、5.77~173.22μg·m L^(-1)(r2=0.999 9)、11.47~344.07μg·m L^(-1)(r2=0.999 8)和2.08~62.04μg·m L^(-1)(r2=0.999 9)质量浓度范围内与峰面积具有良好的线性关系;平均回收率(n=9)分别为100.4%,100.4%,99.4%和99.8%。结论:该方法简便、专属、重复性好,可作为丹灯通脑胶囊的质量控制方法。
Objective: To develop an HPLC method for simultaneous determining the contents of puerarin,scutellarin,salvianolic acid B and tanshinoneⅡA in Dandeng Tongnao Capsules. Methods: The analysis was performed on an Inerstil-ODS-SP column( 4. 6 mm × 250 mm,5 μm) with gradient mobile phase of acetonitrile-0. 1%phosphoric acid solution at a flow rate of 1 m L·min^(-1). The detection wavelength was set at 280 nm and the column temperature was 30 ℃. Results: The linearity ranges of the calibration curves of ferulic acid,salvianolic acid B,puerarin and scutellarin were 3. 85 ~ 115. 41( r2= 0. 999 8),5. 77 ~ 173. 22( r2= 0. 999 9),11. 47 ~ 344. 07( r2= 0. 999 8),and 2. 08 ~ 62. 04 μg·m L^(-1)( r2= 0. 999 9),respectively. The average recoveries( n = 9) were100. 4%,100. 4%,99. 4% and 99. 8%,respectively. Conclusion: The method is convenient,selective and reproducible and can be used for quality control of Dandeng Tongnao Capsules.
