摘要
目的:探讨人乳腺癌细胞MCF-7中mi R-145是否通过调控c-Myc参与肿瘤细胞的生长。方法:化学合成mi R-145-3p和mi R-145-5p并分别转染MCF-7细胞;CCK-8法检测转染后24、48、72 h的细胞增殖情况;RT-PCR检测转染72 h后MCF-7细胞中c-Myc m RNA表达量的变化。结果:与阴性对照组和空白组相比,CCK-8结果显示5p组细胞增殖抑制显著(P<0.05),而3p组抑制效果不明显((P>0.05));RT-PCR结果显示,5p组和3p组细胞中c-Myc m RNA的表达量均下调,且5p组细胞的下调更显著(P<0.05)。结论:乳腺组织中mi R-145负调控c-Myc的表达,可能参与人乳腺癌的发生过程。
Objective:To investigate whether mi R-145 in human breast cancer MCF-7 cells is involved in the growth of MCF-7 cells by regulating c-Myc.Methods:MCF-7 cells were transfected with chemically synthesized mi R-145-3p or mi R-145-5p.Cell Counting Kit-8(CCK-8)assay was used to measure cell proliferation at 24,48,and 72 hours after transfection,and RT-PCR was used to measure the change in the m RNA expression of c-Myc at72 hours after transfection.Results:Compared with the negative control group and blank group,the 5p group had significant inhibition of cell proliferation,while the 3p group did not have marked inhibition,according to CCK-8results.RT-PCR results showed downregulated m RNA expression of c-Myc in both 5p group and 3p group,and the5p group had a significantly greater reduction.Conclusion:mi R-145 significantly downregulates the expression of c-Myc in breast tissue and may thus be involved in the development of human breast cancer.
