摘要
目的:探讨下调microRNA-107(miR-107)表达对人子宫内膜样腺癌细胞系Ishikawa增殖迁移和侵袭能力的影响及可能机制。方法:将miR-107抑制剂(miR-107 inhibitor)瞬时转染Ishikawa细胞,下调miR-107表达。实时荧光定量PCR法(qRT-PCR)检测miR-107表达,CCK-8法检测细胞增殖能力,细胞划痕方法检测细胞迁移能力,Transwell小室法检测细胞侵袭能力,Western blot法检测ER-α受体蛋白表达水平。结果:转染miR-107 inhibitor的Ishikawa细胞中miR-107表达水平显著降低,细胞的增殖迁移和侵袭能力降低,细胞中ER-a受体蛋白表达水平增高。结论:下调miR-107表达可能通过上调ER-α表达抑制Ishikawa细胞的增殖迁移和侵袭能力。
Objective: To investigate the effects of down-regulated expression of hsamiR-107 on the proliferation migration and invasion of Ishikawa cells and its mechanism.Methods: miR-107 inhibitor was transiently transfected into Ishikawa cells to down-regulate expression of miR-107.The expression of miR-107 was detected using quantitative reverse transcription polymerase chain reaction( qRT-PCR). Cellular proliferation,migration and invasion abilities were analyzed by cell counting kit-8( CCK-8) assay,wound healing assay and transwell migration assay,respectively.Western blot was used to detect the expression of ER-α( estrogen receptor alpha).Results: The expression of miR-107 in Ishikawa cells transfected with miR-107 inhibitor was significantly decreased,the proliferation,migration and invasion of cells were decreased,and the expression of ER-a in cells was increased. Conclusions: Down-regulation of miR-107 inhibited the proliferation,migration and invasion of Ishikawa cells by up-regulating the expression of ER-α.
