摘要
目的观察以健脾补肾、调肝活血、温通泻浊为治法组方的中药复方甲荣康对甲状腺功能减退模型大鼠卵巢组织InhBP表达的影响并探讨其机制。方法用他巴唑复制甲状腺功能减退大鼠模型,并分为5组(n=10):模型对照组、西药对照组、甲荣康小、中、大剂量组,另加空白对照组,分别灌胃。药物干预6周后,采用免疫组化方法检测各组大鼠卵巢组织InhBP的表达。结果甲减模型大鼠卵巢组织InhBP表达水平明显升高,甲荣康可显著的降低模型大鼠卵巢组织InhBP表达水平,而且随着剂量的加大,疗效更加显著,明显优于对照组。结论 InhBP的异常表达是甲减合并性腺功能减退的发病机制之一,通过健脾补肾、调肝活血、温通泻浊法对甲减合并性腺功能减退进行早期干预效果明显,甲荣康可能通过降低InhBP在模型大鼠卵巢组织中的异常表达对甲状腺及性腺功能起到保护作用,值得进一步深入研究。
Objective： To observe the effect of TCM compound Jiarongkang on InhBP expression in ovarian tissue from hypothyroidism and hypogonadism rats and discuss its mechanisms,which is originally aimed to nourish spleen,kidney,liver and bone marrow with alleviating diarrhea. Methods： The rat model of hypothyroidism was established by orally administering tapazole and these hypothyroidism rats were divided into 5 groups（ n = 10 each） ： the model control group,the western medicine control group,the Jiarongkang group with low,medium and high doses,additionally the blank control group included. All of medications were given orally. After 6 weeks of intervention,the expression of InhBP in ovarian tissues was detected by immunohistochemistry. Results： The InhBP expression in ovarian tissues from hypothyroidism rats was significantly increased. Jiarongkang significantly reduced the expression of InhBP in ovarian tissues in the hypothyroidism rats. In addition,the therapeutic effect was more significant with the increase of dose,significantly better than that of the other control groups with tapazole administration. Conclusion： Abnormal InhBP expression is one of the mechanisms in hypothyroidism combined with hypogonadism. Early intervention of hypothyroidism plus hypogonadism can be effective by nourishing spleen,kidney,liver and bone marrow with attenuating diarrhea. Jiarongkang possibly reduces the abnormal expression of InhBP in ovary tissues from the model rats,which plays a protective role in thyroid gland and gonadal function,and remains to be studied.
