不同脱蜡剂对实时荧光定量PCR检测石蜡块EGFR基因突变结果的影响 被引量:1

The effects of different dewaxing agents on the detection of EGFR gene mutation in paraffin-embedded tissues specimens by real-time quantitative PCR method

摘要 目的观察不同的脱蜡剂对实时荧光定量PCR检测表皮生长因子受体(EGFR)基因结果判读的影响。方法从2014年11月起,连续6个月收集该院病理科石蜡标本切片68份,均分成A组(二甲苯脱蜡)和B组(松节油脱蜡)进行石蜡块EGFR基因突变检测。结果同一份石蜡切片检测EGFR的6个靶点两组结果均相同。两组同一份标本的内质控(内参)扩增曲线CT值之差均小于0.99;同一份标本,两组检测相同靶点时有效的扩增曲线CT值之差均小于0.99,且结合内质控(内参)CT值分析最终判定两组靶点检测的阳性、阴性结果完全相符。结论采用松节油与二甲苯脱蜡后检测EGFR基因突变的实时荧光定量PCR结果一致,对检测的靶点无影响,松节油可用作分子实验室的脱蜡剂。 Objective To observe the effects of different dewaxing agents on the detection of epidermal growth factor receptor(EGFR)gene mutation in paraffin-embedded tissues specimens by real-time quantitative PCR.Methods From November 2014,68 paraffin-embedded specimens were were consecutively collected from the department of pathology in this hospital for 6 months.All specimens were each treated with different dewaxing agents:the group A was treated with xylene dewaxing,the group B was treated with turpentine dewaxing.Then DNA was extracted by DNA extraction kit and detected by real-time quantitative PCR.ResultsThe results of 6 targets of EGFR in each paraffin-embedded section were consistent for the two groups.For each paraffin-embedded section,the difference of CT value of interval reference amplification curve between the group A and the group B was less than 0.99.For the same targets of each paraffin-embedded section,the difference of CT values of effective amplification curve between the group A and the group B were less than 0.99.Considering the CT value of internal control(internal reference),the positive and negative results of targets in group A were consistent with those in group B.Conclusion The results of epidermal growth factor receptor(EGFR)gene mutation in turpentine dewaxing paraffin-embedded tissues specimens are consistent with those in xylene dewaxing paraffin-embedded tissues specimens by real-time quantitative PCR,without influences on detection results of each target,which indicates turpentine could serve as an alternative dewaxing agent for molecular laboratory.
