摘要 目的研究循环肿瘤细胞(CTCs)富集和鉴定方法。方法分别用羧基磁珠制备EPCAM、CK82种免疫磁珠,分成EPCAM组、CK8组、2种磁珠均等混合检测的EPCAM/CK8组3组,对循环肿瘤细胞进行富集,采用HE染色和免疫细胞化学法鉴定细胞。结果 EPCAM组、CK8组、EPCAM/CK8组检测值分别为12.4±7.3、9.8±4.2、21.6±5.9个肿瘤细胞,检测效率差异有统计学意义(P<0.05)。结论 EPCAM与CK8联合检测可有效提高非小细胞肺癌CTCs检出效率。 Objective To study the enrichment and identification of circulating tumor cells(CTCs).Methods Two kinds of immunomagnetic beads,EPCAM and CK8,were prepared by carboxyl magnetic beads.There were three groups:EPCAM group,CK8 group,and EPCAM/CK8 group in which two kinds of magnetic beads homogeneously mixed for detection.Tumor cells were enriched,and HE staining and immunocytochemical method were used to identify cells.Results The detection values of EPCAM group,CK8 group and EPCAM/CK8 group were 12.4±7.3,9.8±4.2,21.6±5.9 tumor cells,respectively,and the difference in detection efficiency was statistically significant(P〈0.05).Conclusion The combined detection of EPCAM and CK8 could effectively improve the detection efficiency of CTCs in non-small cell lung cancer.
