摘要
透射电镜下观察Caco-2细胞形态,是大多数实验室评价Caco-2细胞单层模型完整性的常用方法,但其对Caco-2细胞表面形态的观察上存在某些不足,特别是在微绒毛的观察上。扫描电镜能更直观清晰地观察到Caco-2细胞单层模型的表面形态结构特征。本文详细描述了在透射电镜和扫描电镜下观察到的Caco-2细胞单层模型的形态结构,并认为将透射电镜下观察到的Caco-2细胞极性、连接复合体以及微绒毛形态,与扫描电镜下观察到的Caco-2细胞形态、大小和排列以及微绒毛长度、密度,两者进行结合与补充,对Caco-2细胞单层模型形态学方面的评价,将会更为全面和完整。
Observation of Caco-2 cell morphology with transmission electron microscopy( TEM) is a common method to assess the integrity of Caco-2 cell monolayer model in most laboratories. However,there are some deficiencies in the observation of the surface morphology of Caco-2 cells with TEM. It can easily lead to the difference of microvilli morphology due to the difference of the cutting plane during the process of embedding and slicing of Caco-2 cell monolayer samples. So,it may affect the accuracy of the results if we only use the morphological changes of microvilli of Caco-2 observed under TEM to evaluate the effect of drugs on Caco-2 cell monolayer model. In contrast,the surface morphological characteristics of Caco-2 model are acquired more directly and clearly by scanning electron microscopy( SEM). In this paper,we explicitly described the structure of Caco-2 cell monolayer model with TEM and SEM respectively. And,we think we should combine cell polarity,cell junction complex and microvilli of Caco-2 observed under TEM with information about shape,size,arrangement,density and length of microvilli of Caco-2 observed under SEM. It will be more comprehensive to evaluate the morphology of Caco-2 cell monolayer model with the combination of these two methods.
