摘要
以D-半乳糖制衰老模型小鼠为研究对象,评价深海鲣鱼鱼油对衰老模型小鼠抗衰老的作用。以健康育龄ICR小鼠为试验对象,随机分成6组,每组8只,即为空白对照组;鲣鱼鱼油制剂组:低、中、高剂量组;D-半乳糖模型组;阳性对照组。采用皮下注射方式,空白对照组小鼠每只每天注射0.9%的生理盐水,其余实验组注射浓度为的4%D-半乳糖(1 000mg/kg),同时,鲣鱼鱼油制剂组分别每天灌胃10 m L/kg bw不同质量浓度的鱼油制剂(20,50,100 mg/m L),阳性组灌胃市售鱼油(100 mg/kg),空白对照组和模型组灌胃等体积生理盐水,持续6周。随后检测各组小鼠肝脾指数,肝脏组织和血清的GSH-Px,T-SOD,CAT活性,MDA含量等指标,制备肝脏切片,观察小鼠肝脏组织的病理变化情况。结果表明,鲣鱼鱼油高剂量组和空白对照组的肝脾指数均呈明显下降趋势;模型组小鼠血清和肝组织GSH-Px,T-SOD,CAT活性明显降低,MDA含量显著升高,与空白组比较有显著性差异,鲣鱼鱼油各剂量组均能提高血清和肝组织中GSH-Px,T-SOD,CAT活性并降低MDA含量;通过切片观察,深海鲣鱼鱼油可缓解衰老过程中肝脏的损伤,并且高剂量组的效果最为明显。
D-galactose induced aging model mice were used as research objects to evaluate the anti-aging effect of deep-sea Katsuwonus pelamis oil on aging model mice. The healthy reproductive age ICR mice were randomly divided into 6 groups(n=8), which were blank control group, skipjack tuna oil group with low,middle and high dose of catfish fish oil, the model group of D-galactose and the positive control group. Subcu-taneous injection, the control group mice were injected 0.9% saline every day, while the remaining experimental group was injected with 4% D-galactose(1 000 mg/kg) at the same time, the same time,(20, 50, 100 mg/m L)with different concentrations of fish oil(10 m L/kg bw). The positive group was fed with fish oil(100 mg/kg) on the market and the blank control group and model group were given gavage with equal volume of normal saline for 6 weeks. The liver tissues and serum of mice were then tested for GSH-Px, T-SOD, CAT activity, MDA content and other indicators, liver sections were prepared and pathological changes of liver tissues were observed.The liver and spleen index of the high-dose group and the blank control group were significantly decreased; The activities of GSH-Px, T-SOD and CAT in the serum and liver tissue of the model group were significantly decreased, MDA content was significantly increased, compared with the control group, the levels of GSH-Px, TSOD and CAT in serum and liver tissue were significantly decreased. By slicing observation, deep-sea K.pelamis oil can alleviate the liver damage during aging, and the effect of high-dose group is the most obvious.
