产后抑郁大鼠脑前额叶皮层、海马区DA及其代谢产物、受体动态变化及参芪解郁方的干预作用 被引量:9

Intervention mechanism of Shenqi Jieyu Formula on dynamic changes of DA and its metabolite, receptors in prefrontal cortex and hippocampus in rats with postpartum depression

摘要 目的:从脑前额叶皮层、海马DA、DOPAC含量及D2R表达变化角度,探讨具有补益心脾作用参芪解郁方对产后抑郁症(PPD)的干预作用。方法:选用雌性2~3月龄SD大鼠,随机分为正常组、假手术组、模型组、中药组、西药组,每组设1、2、4、6周观察时点。选取脑前额叶皮层、海马区域,采用高效液相法测定DA及其代谢产物DOPAC含量,免疫组化法检测D2R表达。结果:模型组各时点前额叶皮层、海马DA、DOPAC含量均较正常组减少(P<0.01)。中药组4、6周及西药组2、4、6周前额叶皮质,中药组1、6周及西药组1周海马DA含量均较模型组同期升高(P<0.05,P<0.01);中、西药组2、4、6周前额叶皮层DOPAC含量较模型组同期升高(P<0.05,P<0.01),各时点海马DOPAC含量均较模型组无明显差异。模型组各时点前额叶皮层、海马D2R表达均较正常增多(P<0.01)。中药组各时点及西药组1、4、6周前额叶皮层,中药组各时点及西药组4、6周海马D2R表达均较模型组减少(P<0.05,P<0.01)。结论:参芪解郁方能够明显改善PPD大鼠脑前额皮质、海马DA的功能低下,调节DOPAC的紊乱失衡状态及D2R表达障碍,其效应发挥可能与该方对单胺递质DA及其代谢产物、受体的稳态调节途径相关。 Objective: In this study, Shenqi Jieyu Formula with the effect of tonifying heart and spleen was used to treat postpartum depression (PPD) model animals, and changes on contents of DA, DOPAC and expressions of D2R in prefrontal cortex, hippocampus were tested. To explore the intervening mechanism of Chinese medical therapeutic method of tonifying heart and spleen. Methods: Female SD rats, 2 to 3 months of age, were randomly divided into groups of normal (N), sham operation (SO), model (M), Chinese medicine (CM), western medicine (WM). All the rats were observed at l, 2, 4, 6w respectively after preparation, with behaviors tests at each time points above, then surveying contents of DA and its metabolite DOPAC with HPLC, as well as expressions of D2R with IHC in prefrontal cortex and hippocampus. Results: Compared with N group, contents of DA and DOPAC in prefrontal cortex and hippocampus in M group at each time point were all decreased (P〈0.01). Compared with M group, contents of DA group in prefrnntal cortex in CM group at 4, 6w and WM group at 2, 4, 6w, as well as in hippocampus in CM group at 1, 6w and WM group at lw were all increased (P〈0.05, P〈0.01). Compared with M group, the contents of DOPAC in prefrontal cortex in CM group and WM group at 2, 4, 6w were all increased (P〈0.05, P〈0.01), meanwhile, that contents in hippocampus in CM group and WM group at each time point with no significant changes. Compared with N group, expressions of D2R in prefrontal cortex and bippocampus in M group at each time point were both enhanced (P〈0.01). Compared with M group, the expressions of D2R in prefrontal cortex in CM group at each time point and WM group at 1, 4, 6w, as well as, that expressions in hippocampus in CM group at each time point and WM group at 4, 6w were all decreased (P〈0.05, P〈0.01). Conclusion: Shenqi Jieyu Formula could remarkably improve the hypofunction of DA in prefrontal cortex and hippocampus of PPD rats, as well as, regulating the disorder of DOPAC and imbalance of D2R expressions. All positive effects above may be exerted the effect on steady-state regulation pattern for monoarnine transmitter DA, as well its metabolites and receptors.
