顶空固相微萃取-气相色谱-质谱联用法检测黄芪挥发油成分 被引量:9

Analysis of chemical compounds of essential oils of Astragalus by SPME-GC-MS

摘要 目的:利用顶空固相微萃取-气相色谱-质谱连用技术检测黄芪挥发油的化学成分,为黄芪的进一步开发和利用提供理论依据。方法:采用顶空固相微萃取技术对黄芪的挥发油成分进行富集,应用气相色谱-质谱联用法对检测出的化学成分进行分离和鉴定,采用面积归一法确定各成分的相对含量。结果:共检出挥发性成分65种,鉴定出其中的62个组分,占挥发油总量的97.70%。主要包括萜烯类和多环芳烃化合物;含量较高的分别是2-戊基呋喃(11.14%)、醋酸(10.69%)、姜黄烯(6.06%)、己腈(5.15%)、α-胡椒烯(3.91%)、椰子醛(3.69%)、羊油酸(3.48%)、正庚酸(3.26%)等。结论:黄芪挥发油化学成分丰富,具有很强的药用价值。顶空固相微萃取-气相色谱-质谱联用能全面快速的获得其组成信息,可应用于黄芪挥发油的快速分析。 Objective: To provide the theoretical basis for the further development and utilization of Astragalus membranaceus. The chemical constituents of volatile oil from Astragalus membranaceus were determined by solid phase microextraction-gas chromatography-mass spectrometry (SPME-GC-MS). Methods: The essential oils were extracted by headspace solid phase micro-extraction (HS-SPME) then separated and identified by gas chromatography-mass spectrometry (GC- MS). The relative content of each component was determined by area normalization. Results: Sixty-five compounds were detected from essential oils of tassel of carrot, and sixty-two of them were identified, which account for 97.70% of total flow area, mainly included terpene and polycyclic aromatic hydrocarbon compounds; the main constituents of the oil were 2-pentylfuran (11.14%), Acetic acid (10.69%), Curcumene (6.06%), Hexanenitfile (5.15%), a -Copaene (3.91%), Prunolide (3.69%), Caproic acid (3.48%), oenanthic acid (3.26%). Conclusion: There are abundant chemical constituents in the Volatile oil of Astragalus membranaceus, which has a strong medicinal value. SPME-GC-MS can get the information of its composition in a full and rapid way. It can be used for the rapid analysis of volatile oil from Astragalus membranaceus.
