摘要
为探索茖葱种子萌发条件及其对PEG-8000处理的响应,同时测定不同生长调节剂配方对茖葱不定芽分化的影响。以前一年和当年茖葱种子为材料,采用培养皿滤纸卷法进行种子萌发试验;以茖葱鳞茎为外植体进行不定芽诱导分化。结果表明,10%(w)的PEG-8000处理可明显促进前一年茖葱陈种子发芽,发芽率94.00%、发芽指数0.89。随PEG-8000质量分数的增加,茖葱种子的发芽率、发芽势及发芽指数有所降低;低温层积50 d及PEG-8000浸种能显著促进茖葱当年新种子萌发,沙藏层积50 d,再用200 mg·L^(-1)PEG-8000浸种4 h,发芽启动时间及发芽高峰期提前最多,发芽持续时间缩短10 d左右,与CK相比,发芽势、发芽率及发芽指数分别提高了475.00%、65.52%及93.75%,对促进茖葱种子发芽的效果最好,同时PEG-8000浸种能显著减少种子层积后的烂种数。筛选出MS+6-BA 1.5 mg·L^(-1)+2,4-D 1.0 mg·L^(-1)+KT 0.1 mg·L^(-1)为供试方案中的最优培养基配方,分化率为56.56%。
In order to explore the germination conditions of the seeds and the response to PEG-8000 treatment, and the effect of different plant growth regulator combinations on Allium victorialis L. adventitious buds induction, Allium victorialis L. seeds of last year and the new one were taken as materials. The seed germination test was carried out with culture dish filter paper. Bulbs of Allium victorialis L. were used as explants for adventitious buds inducted differentiation. The results indicated that the 10% PEG-8000 treatment can obviously promote the germination of old seed. The germination rate was 94.00% and the germination index was 0.89. The germination rate, germination potential and germination index were decreased with the increasing the concentration of PEG-8000. The cold stratification of 50 days and PEG-8000 immersion can significantly promote the germination of the new seeds.The immersion for 4h with 200 mg·L^(-1) PEG-8000 after 50 days of sand accumulation layer, can make the start time and the peak of germination come earlier. The period of germination was reduced by 10 days, compared with CK, germination potential, germination rate and germination index increased by475.00%, 65.52% and 93.75% respectively, the promotion effect of seed germination was the best. At the same time,PEG-8000 immersion can significantly decreased the number of decomposed seeds after lamination. The formula of MS+6-BA 1.5 mg·L^(-1)+ 2,4-D 1.0 mg·L^(-1)+KT 0.1 mg·L^(-1) was selected as the optimal medium in the test, and the differentiation rate was 56.56%.
