摘要
目的探讨不同浓度的橄榄苦苷在不同作用时间对卵巢癌SKOV3细胞增殖的影响。方法 SKOV3卵巢癌细胞传代培养后接种于3块96孔板,每板实验组细胞用100、200、400、800μg/m L的橄榄苦苷培养液,对照组细胞用Mc Coy's5A完全培养基。CCK-8检测法测定24、48、72 h等3个时间点不同药物浓度的SKOV3细胞增殖情况。结果相同作用时间下,与对照组比较,100、200、400、800μg/m L的橄榄苦苷浓度对SKOV3细胞的增殖均有抑制作用,细胞生长A值随着药物浓度增大而减小(P<0.05);在100、200、400μg/m L的药物浓度下,细胞生长A值随着作用时间延长而增大(P<0.05),当药物浓度达800μg/m L时,细胞生长迟缓,至72h细胞A值较24、48h差异有统计学意义(P<0.05);抑制率随着药物浓度增大而增大(P<0.05),与作用时间差异无统计学意义(P>0.05)。结论橄榄苦苷对卵巢癌细胞SKOV3的增殖有抑制作用,具体机制尚待后期实验研究。
Objective To investigate the effects of different concentration of oleuropein on proliferation of ovarian cancer cell SKOV3 in different time. Methods SKOV3 ovarian cancer cells were inoculated in 3 blocks of 96 well plate. Cells of each trial group were treated with 100,200,400,800 μg/m L oleuropein and vehicle,respectively. The proliferation of SKOV3 cells was determined by using CCK-8 assays after the treatment with indicated concentration of oleuropein for 24,48,and 72 h. Results 100,200,400,and800 μg/m L oleuropein significantly inhibited the proliferation of SKOV3 cells. Optical density( OD) was decreased accompanying with the dose increase of oleuropein( P〈0. 05). ODs of SKOV3 cells treated with 100,200,and 400 μg/m L oleuropein were increased for the indicated incubation time( P〈0. 05). The proliferation of cells treated with 800 μg/m L oleuropein for 72 h was retarded compared with those for 24 h and 48 h( P〈0. 05). The inhibition rate of cell proliferation was increased with the dose increase of oleuropein treatment( P〈0. 05). However,there was no significant correlation with the incubation time( P〉0. 05). Conclusion Oleuropein could inhibit the proliferation of ovarian cancer cell SKOV3,the specific mechanisms needs further experimental study.
